Design, Synthesis, and Vasorelaxant and Platelet Antiaggregatory Activities of Coumarin—Resveratrol Hybrids.

Abstract: Hybrids. -The coumarin-resveratrol hybrid (IV) is accessible either by a Pechmann procedure [cf. (I)+(II)→(III)] or by a Perkin reaction. The latter allows also the synthesis of analogue (VIII). The new molecules show remarkable vasorelaxant effects and significant human platelet antiaggregatory activity. They reveal a pharmacological profile similar or even better compared to that of t-RESV. -(VILAR*, S.; QUEZADA, E.; SANTANA, L.; URIARTE, E.; YANEZ, M.; FRAIZ, N.; ALCAIDE, C.; CANO, E.; ORALLO, F.; Bioorg. “…1 A wide variety of bioactivities of these heterocyclic compounds have been revealed, including antimicrobial, 2 antioxidant, 3 antinocipeptive, 4 antitumour, 5 anti-inflammatory, 6 antiasthmatic, 7 antiviral, 8 and vasorelexant. 9 Natural coumarins such as geiparvarin 10 and properly modified natural coumarins, have also been characterised as potent and selective monoamine oxidase (MAO-B and MAO-A) isoforms inhibitors. 11 Reactive oxygen species (ROS) are highly reactive molecules that originate mainly from cell metabolism.…”

“…Resveratrol has been reported to possess a wide range of pharmacological effects, including anti-inflammatory, anticarcinogenic, antioxidant, estrogenic, antiplatelet, and antimicrobial properties. [1][2][3][4][5][6][7] Resveratrol in red wine has been suggested to be responsible for the "French paradox", decreased risk for coronary artery disease in France.…”
