Deoxyribonucleic Acid Homology Among Bacillus polymyxa, Bacillus macerans, Bacillus azotofixans, and Other Nitrogen-Fixing Bacillus Strains

Abstract: Deoxyribonucleic acid homology experiments were performed among representative strains of Bacillus polymyxa, Bacillus macerans, and Bacillus azotofixans and other nitrogen-fixing Bacillus strains identified as B. polymyxa-like. All B . azotofixans strains showed less than 20% homology when B . polymyxa or B . macerans was used as a probe. The range of homology among B. azotofixans strains (strain P3L-5T [T = type strain] was used as the probe) varied from 54 to loo%, indicating that these strains comprise a re… Show more

“…To determine genomic relatedness, the filter hybridization method was performed as described by Seldin & Dubnau (1985). Probe labelling was conducted by using the nonradioactive DIG-High prime system (Roche); hybridized DNA was visualized using the DIG luminescent detection kit (Roche).…”

“…The G+C content of genomic DNA was determined by HPLC (by the Laboratorium voor Microbiologie, Ghent, Belgium) as described previously (Mesbah et al, 1989). To determine genomic relatedness, the filter hybridization method was performed according to Seldin & Dubnau (1985). Probe labelling was conducted by using the non-radioactive DIGHigh prime system (Roche); hybridized DNA was visualized using the DIG luminescent detection kit (Roche).…”

“…Stability of relationships was assessed by performing bootstrap analyses of the neighbour-joining data, based on 1000 resamplings. The nucleotide sequences determined have been deposited in GenBank under the accession numbers shown in Table 1. To determine genomic relatedness, the filter hybridization method was performed according to Seldin & Dubnau (1985). Native DNA was labelled with digoxigenin-11-dUTP by DIG-High Prime (Roche).…”
