“…Our findings also support Girdler et al (24) suggestion that neuroactive steroids are mainly synthesized in the adrenals and that a history of depression may be associated with persistent adrenal suppression (24). Indeed, we show that the DHEA to CT ratio was significantly lower in depressed mothers compared to controls; it seems that DHEA secretion was even more sensitive than CT to allostatic pressures in depressed mothers, a finding that may be of interest when developing new therapeutic strategies for chronic maternal depression (9,12). There is some evidence that the beneficial effects of these agents are mediated by the regulation of the HPA axis (48).…”

“…Furthermore, these hormones seem to have some therapeutic value (11). In a recent meta-analysis, there seem to be a significant treatment effect for these hormones in depression when compared to placebo (12) and DHEA and DHEA-s were found to influence basic electrophysiological processes. An interest finding in this regard is that depression is accompanied by an attenuated DHEA and DHEA-S response to acute psychosocial stress, which may help elucidate the relationship between the HPA axis and stress (13).…”

“…Notably, neurosteroids with Sigma1R agonist properties demonstrate antidepressant-like activity [28,105,108,116,117], while progesterone prevents their effects [28,116]. DHEA has been shown to possess antidepressant effects when administered to patients with depressive disorders [118].…”

“…Another RCT applying a 1-year DHEA treatment did also not reveal mood alleviating effects of DHEA in 110 men aged 55 to 85 years [88]. Additionally, there has been a recent yet somewhat questionable meta-analysis which included RCTs on DHEA supplementation suggesting beneficial effects of DHEA in depression [93]. However, based on these inconsistencies and the general shortage of empirical evidence, DHEA is not being considered as a potential antidepressant treatment in men.…”
