This study aimed to illustrate the use of D-optimal mixture design (DOMD) for optimization of an enhancer containing Dapsone niosomal formula for acne topical treatment. Mixture components (MixCs) studied were: Span 20, Cholesterol, and Cremophor RH. Different responses were measured. Optimized formula (OF) was selected to minimize particle size and maximize absolute zeta potential and entrapment efficiency. Optimized formula gel (OF-gel) was prepared and characterized. OF-gel in vivo skin penetration using confocal laser scanning microscopy and activity against Cutibacterium acnes in acne mouse model were studied. Based on DOMD results analysis, adequate models were derived. Piepel and contour plots were plotted accordingly to explain how alteration in MixCs L-pseudo values affected studied responses and regions for different responses’ values. The OF had suitable predicted responses which were in good correlation with the actually measured ones. The OF-gel showed suitable characterization and in vivo skin penetration up to the dermis layer. In vivo acne mouse-model showed that OF-gel-treated group (OF-gel-T-gp) had significantly better recovery (healing) criteria than untreated (UT-gp) and Aknemycin ® -treated (A-T-gp) groups. This was evident in significantly higher reduction of inflammation percent observed in OF-gel-T-gp than both UT-gp and A-T-gp. Better healing in OF-gel-T-gp compared with other groups was also verified by histopathological examination. Moreover, OF-gel-T-gp and A-T-gp bacterial loads were non-significantly different from each other but significantly lower than UT-gp. Thus, DOMD was an adequate statistical tool for optimization of an appropriate enhancer containing Dapsone niosomal formula that proved to be promising for topical treatment of acne.

Emerging Technologies to Target Drug Delivery to the Skin – the Role of Crystals and Carrier-Based Systems in the Case Study of Dapsone

Dapsone (DAP) is a long-established molecule that remains a promising therapeutic agent for various diseases mainly because it combines antimicrobial and antiinflammatory activities. Its oral application, however, is limited by the dose-dependent hematological side effects that may rise from systemic exposure. As an alternative to overcome this limitation, the administration of DAP to the skin has witnessed prominent interest in the past 20 years, particularly when applied to the treatment of dermatological disorders. In this review, all technological strategies proposed to the topical delivery of DAP are presented. Most of the reported studies have been devoted to the clinical use and safety of a gel formulation containing both solubilized and microcrystalline drug, however, the technological characteristics of such preparation are still missing. In parallel, the incorporation of DAP into vesicular and particulate carriers (e.g. nano-and microemulsions, niosomes, invasomes, bilosomes, cubosomes, solid lipid nanoparticles, nanostructured lipid carriers, polymeric nanocapsules and polymer-lipid-polymer hybrid nanoparticles) appears to be an alternative to provide greater drug release control, enhanced drug solubilization and follicular targeting. Indeed, the main application of DAP topical formulations reported in the literature was the treatment of acne vulgaris, a disease located in the hair follicle. Other diseases affecting different regions of the skin (e.g. cutaneous lupus erythematosus and cutaneous leishmaniasis), however, may also benefit from a topical therapeutic regimen containing DAP. Therefore, the investigation of appendageal route in comparison to passive transmembrane diffusion as a function of targeted disease, as well as pharmacokinetic studies, are perspectives highlighted herein. Such studies may drive future efforts towards the rational development of safe and effective technologies to deliver DAP to the skin.

Recent research in drug delivery has significantly advanced our knowledge of liposome and niosome‐based gels, contributing to the dynamic landscape of pharmaceutical advancements. The incorporation of these vesicular carriers into gel formulations enhanced stability, while prolonging shelf‐life and improving the bioavailability of encapsulated molecules. The synergy between liposomes/niosomes and gel matrices, facilitated by a robust gel structure, provided a protective environment, ensuring sustained drug release and prolonged stability. Liposome‐based gels proved to be versatile carriers for various therapeutic applications, demonstrating efficiency in solubilizing poorly soluble drugs and acting as potent penetration enhancers for optimal skin delivery. Today, the incorporation of liposomes in gels serves as local depots, thus being a fundamental condition for sustained and controlled drug release to optimize patient's outcomes. Insights into niosome‐based gels reveal their potential in overcoming conventional drug delivery challenges, positioning them as promising platforms for controlled and targeted drug release due to enhanced stability. Recent investigations highlight the adaptability of niosomal gels in encapsulating both hydrophobic and hydrophilic drugs, ensuring superior stability and sustained release. The tunable properties of niosomal gels also contributed to optimize bioadhesion and permeation across biological membranes. The primary objective of this work is to present the cutting‐edge findings in liposome and niosome‐based gel research, highlighting their path in modern drug delivery. These innovative solutions offer more efficient and targeted therapeutic interventions, addressing the ever‐evolving landscape of medicine.
