Oroxylum indicum (L.) Kurz, a medicinal plant, shows numerous pharmacological properties which may be attributed to the bioactive compounds produced by O. indicum or due to associated endophytes. In the present study, leaf of O. indicum was evaluated for the presence of associated fungal endophytes, and antioxidant and cytotoxic activities of bioactive compounds produced from them. Using culture-dependent approach, eight fungal endophytes belonging to five different genera were identified. Two endophytes Daldinia eschscholtzii and Ectophoma multirostrata have been reported for the first time from the leaf of O. indicum plant. High-performance thin-layer chromatography (HPTLC) of ethyl acetate (EA) extract of isolated fungal endophytes showed a distinct fingerprinting profile in EA extract of Colletotrichum gloeosporioides. Among identified endophytes, EA extract of C. gloeosporioides showed significant antioxidant activity against DPPH free radical, superoxide anion radical, nitric oxide radical and hydroxyl radical with EC50 values of 22.24±1.302 μg/mL, 67.46±0.576 μg/mL, 80.10±0.706 μg/mL and 61.55±1.360 μg/mL, respectively. EA extract of C. gloeosporioides exhibited potential cytotoxicity against HCT116, HeLa and HepG2 cancer cell lines with IC50 values of 76.59 μg/mL, 176.20 μg/mL and 1750.70 μg/mL, respectively. A comparative HPTLC fingerprinting and the antioxidant activity of C. gloeosporioides associated with two different hosts (leaf of O. indicum and dead twigs of other plant) showed that C. gloeosporioides produces bioactive compounds in a host-dependent manner.

Human life has been significantly impacted by the creation and spread of novel species of antibiotic-resistant bacteria and virus strains that are difficult to manage. Scientists and researchers have recently been motivated to seek out alternatives and other sources of safe and ecologically friendly active chemicals that have a powerful and effective effect against a wide variety of pathogenic bacteria as a result of all these hazards and problems. In this review, endophytic fungi and their bioactive compounds and biomedical applications were discussed. Endophytes, a new category of microbial source that can produce a variety of biological components, have major values for study and broad prospects for development. Recently, endophytic fungi have received much attention as a source for new bioactive compounds. In addition, the variety of natural active compounds generated by endophytes is due to the close biological relationship between endophytes and their host plants. The bioactive compounds separated from endophytes are usually classified as steroids, xanthones, terpenoids, isocoumarins, phenols, tetralones, benzopyranones and enniatines. Moreover, this review discusses enhancement methods of secondary metabolites production by fungal endophytes which include optimization methods, co-culture method, chemical epigenetic modification and molecular-based approaches. Furthermore, this review deals with different medical applications of bioactive compounds such as antimicrobial, antiviral, antioxidant and anticancer activities in the last 3 years.

A multitude of plants from the Brazilian savanna are known for their medicinal properties. Many plants contain endophytic fungi, which lead to the production of bioactive compounds by both the fungi and their hosts. This study investigated the bioprospecting of endophytic fungi recovered from the leaves of Palicourea rigida, a native medicinal plant of the Brazilian savanna. Four fungal taxa (Colletotrichum sp. SXS649, Pestalotiopsis sp. SXS650, the order Botryosphaeriales SXS651, and Diaporthe sp. SXS652) were recovered. The phenolic, flavonoid, extracellular degrading enzymes (amylase, cellulase, protease, and tannase) and antioxidant activity of these taxa were determined. Evaluation of the antimicrobial activity showed that the Botryosphaeriales SXS651 extract displays a minimum inhibitory concentration (MIC) of 23.20 mg mL‒1 against Staphylococcus epidermidis and Pseudomonas aeruginosa, and the Diaporthe sp. SXS652 extract exhibited an MIC of 27.00 mg mL−1 against Escherichia coli. The Colletotrichum sp. SXS649 isolate inhibited tumors in potato discs by 69% at a concentration of 9.70 mg mL‒1. All isolates had potential bioremediation criteria against soil contaminated with soybean oil, as proved by a high percentage of germination of Lactuca sativa and a reduction in phytotoxicity. Furthermore, the taxa under investigation demonstrated antagonistic action to phytopathogenic fungi, namely, Aspergillus niger, Inonotus rickii, Pestalotiopsis mangiferae, and Coniophora puteana, with an inhibition range between 34.2% and 76.9%. The preliminary toxicity assessment showed that all isolates possessed an LC50 of less than 100 mg mL‒1 to the microcrustacean Artemia salina. These results indicate that the endophytic fungi of the Brazilian savanna are promising candidates for biotechnological and industrial applications and, in agricultural applications, for the biological control of phytopathogenic fungi.
