Ipomea asarifolia, Commelina diffusa, Acalypha ciliata and Eleusine indica are medicinal plants with anthelmintic effects traditionally used in pig breeding in the Republic of Benin. However, data on their chemical composition and toxicity are not sufficient. The objective of this study was to evaluate the phytochemistry and larval toxicity of I. asarifolia, C. diffusa, A. ciliata and E. indica, used in Benin. Aqueous and hydro-ethanolic extracts of the plants were obtained by maceration of the powders in solvents for 48h. Phytochemical screening of the different secondary metabolites present in the plants was performed on the basis of staining and/or precipitation reactions on the powders. Larval toxicity was performed on Artemia salina larvae obtained after the hatching of the crustacean eggs. The determination of the number of dead larvae according to the concentrations used allowed to establish the lethal concentrations 50. From the results obtained, it was found that the different plants contained several secondary metabolites including flavonoids, mucilages, reducing compounds, anthocyanins, anthraquinones, leucoanthocyanins, catechic tannins and gall tannins. Alkaloids were not identified in any of the plants. Toxicologically, all the Lethal Concentrations 50 are higher than 0.1mg/mL, reflecting no toxic character of the extracts at the tested doses. In sum, the plants studied are of interest in traditional medicine because of their interesting chemical composition and the absence of cytotoxic effect. These results justify their use in traditional medicine.

Commelina diffusa Burm. f. is a member of Commelinaceae family, which is widely grown in crop land and treated as a weed. This plant has several important medicinal properties which have not been studied extensively. In this study, the crude powder of C. diffusa whole plant was extracted with 95% ethanol and different solvent fractions (n-hexane, chloroform and methanol) were prepared from the crude extract by solvent-solvent partitioning. All these plant samples were subjected to bioassays for evaluating the antioxidant, central analgesic, peripheral analgesic and CNS depressant activities. The crude extract and its methanol soluble fraction showed mild free DPPH scavenging property with IC50 values of 98.49- and 84.77-μg/ml, respectively as compared to the standard ascorbic acid (IC50 = 2.67 μg/ml). In the analgesic activity test, the n-hexane fraction of C. diffusa at doses of 200- and 400-mg/kg body weight exhibited significant (p<0.05) central analgesic activity (tail flick test) in mice. Similarly, all the test samples showed statistically significant (p<0.05) reduction in abdominal writhing induced by acetic acid. C. diffusa showed significant CNS depressant activity which was measured by ‘open field test’ and ‘hole cross test’. Considering the potential bioactivities, the plant materials can be further studied elaborately to explore the activities of the purified compounds to aid in future drug development. Dhaka Univ. J. Background: Endophytic fungi are able to produce many secondary metabolites and thus their availability and biological activity created new horizons for different pharmaceutical and agricultural approaches. Objective: For the ethnobotanical antiquity of Commelina diffusa in Bangladesh, this study is aimed at evaluating the endophytic fungal collection of Commelina diffusa and their bioactive potential. Methods: Endophytic fungi were isolated by the surface sterilization technique and identified using the microscopic procedure. Isolated fungal strains were solvent extracted. Disc diffusion method, DPPH free radical scavenging assay, brine shrimp lethality bioassay had been used for the determination of antibacterial, antioxidant, cytotoxic activities respectively. Results: Eight fungal strains were isolated and six of them were identified up to the genus level as Fusarium sp. (CDLE 1, CDRE 1, CDRE 2, CDRE 3, CDBE 1) and Alternaria sp. (CDBE 3) according to morphological and microscopical characters. Three of the eight isolated endophytes i.e., CDBE 2, CDRE 1 and CDRE 2 showed potential activity against A. niger in comparison with ketoconazole (30 μg/disc). Ethyl acetate extract CDBE 3 fungal extract demonstrated significant antioxidant and cytotoxic activities. Conclusion: The present study has proven that Commelina diffusa may be a rich source of medicinally important endophytic fungi and our findings may form a basis for further studies on endophytic fungi from medicinal plants for use in medicine, industry and agriculture.
