Cytokines Modulate Expression of Cell-Membrane Complement Inhibitory Proteins in Human Lung Cancer Cell Lines

Abstract: Human lung cancers overexpress several cell-membrane complement inhibitory proteins (CIP). These complement inhibitory proteins are membrane cofactor protein (CD46), decay-accelerating factor (DAF; CD55), and CD59 (protectin). These cell-membrane proteins have a wide normal tissue distribution, are known to protect normal host cells from homologous complement-mediated lysis, and are thought to facilitate tumor escape from immunosurveillance. To study whether proinflammatory cytokines that are involved in cance… Show more

“…For example, CD46 synthesis on human lung cancer cell lines is increased by IL-1␣ and TNF. 42 CD55 expression was found to be upregulated by TNF, IL-1␤, IL-4 and PMA in HUVEC. 43 Bjørge et al 44,45 reported that TNF and IL-1␤ enhanced expression of both CD55 and CD59 in a colon adenocarcinoma cell line, whereas CD46 expression was influenced only by IL-1␤.…”

“…Based on our recent studies,14 it appears that the expression of DAF in human lung cancer cell lines responds somewhat differently to cytokine stimulation than in human bronchial epithelium. DAF expression in ChaGo K-1 lung cancer cells was more sensitive and more responsive to TNF-α stimulation than in BEAS-2B cells.…”

“…77 Expression of mCRPs is modulated by cytokines. 78 For example, IL-1b downregulated the expression of CD46 and CD59, whereas IL-4 only affected CD46 expression. Cytokine-induced downregulation of mCRPs enhanced CDC of renal tumor cell lines in vitro.…”
