CYLD: a tumor suppressor deubiquitinase regulating NF-κB activation and diverse biological processes

Abstract: Protein ubiquitination is a reversible reaction, in which the ubiquitin chains are deconjugated by a family of deubiquitinases (DUBs). The presence of a large number of DUBs suggests that they likely possess certain levels of substrate selectivity and functional specificity. Indeed, recent studies show that a tumor suppressor DUB, cylindromatosis (CYLD), has a predominant role in the regulation of NF-κB, a transcription factor that promotes cell survival and oncogenesis. NF-κB activation involves attachment of… Show more

“…CYLD, a DUB mutated in familial cylindromatosis, was shown to specifically target Lys63-based ubiquitin chains on TRAF2 and TRAF6, and consequently has an inhibitory effect on the NF-kB pathway. 78 In addition, TRAF6 might also be regulated by A20, another Lys63-linked ubiquitin chain-specific DUB that was shown to deubiquitinate TRAF6, among other substrates in the NF-kB pathway. 79 Importantly, A20 was found to be a double function enzyme -a DUB and a ligase.…”

“…38 binds to the genomic region encompassing 16q12 TAO-associated SNP (rs1861760) was 394 described to inhibit NFkB1 activity. 42 Recently, CYLD has been shown to regulate lung 395 fibrosis in mice by inhibiting transforming growth factor-β-signaling (TGF-β) through a 396 decrease of SMAD3 protein stability.…”

“…Cylindromatosis (CYLD) is a tumor suppressor protein harboring deubiquitinase activity that primarily removes lysine 63-linked polyubiquitin chains from a number of signaling components [9]. CYLD also contains cytoskeleton-associated protein glycine-rich (CAP-Gly) domains that mediate its interaction with microtubules and histone deacetylase 6 (HDAC6) [10][11][12].…”
