Cycloastragenol Is a Potent Telomerase Activator in Neuronal Cells: Implications for Depression Management

Abstract: Cycloastragenol (CAG) is an aglycone of astragaloside IV. It was first identified when screening Astragalus membranaceus extracts for active ingredients with antiaging properties. The present study demonstrates that CAG stimulates telomerase activity and cell proliferation in human neonatal keratinocytes. In particular, CAG promotes scratch wound closure of human neonatal keratinocyte monolayers in vitro. The distinct telomerase-activating property of CAG prompted evaluation of its potential application in the… Show more

“…However, in Gur's study, cycloastragenol (CA) is more effective than AS‐IV for wound treatment . According to latest research, CA could improve the activity of telomerase and the proliferation of human neonatal keratinocytes and induce the cAMP response element binding (CREB) activation followed by tert and bcl2 expression . AS‐IV and CA also modulate the cellular effects via activation of src/MEK/ERK signaling pathway in multiple cell lines of vitals .…”

“…Certain phytochemicals such as Astralagus membranaceus root are reported for their telomerase activating capacities [95]. Since telomerase activation plays a significant role in cancer development such food supplements have been debated intensely and are still discussed for their potential cancer risk.…”

“…Further, expression of molecules downstream from the JAK/STAT signaling pathway was also increased, suggesting that CAG activates telomerase through the JAK/STAT signaling pathway (9). In addition, CAG-mediated activation of telomerase is associated with the cAMP response element-binding protein (CREB) (3). The above preliminary studies of the mechanisms associated with CAG demonstrate that it activates telomerase and is related to the CREB, MAPK, and JAK/STAT pathways; however, further investigation is needed to comprehensively understand the mechanisms.…”
