Abstract: Although growth factors and chemokines elicit different overall effects on cells-growth and chemotaxis, respectively-and activate distinct classes of cell-surface receptors, nonetheless, they trigger similar cellular activities and signaling pathways. The growth factor M-CSF and the chemokine CXCL12 both stimulate the endocytic process of macropinocytosis, and both activate the mechanistic target of rapamycin complex 1 (mTORC1), a protein complex that regulates cell metabolism. Recent studies of signaling by M… Show more

“…Interestingly, Swanson and colleagues have recently shown that macropinosomes can serve as signaling scaffolds for the propagation of GPCR signaling to PI3K [22]. Given that the expression of activated Rac1 stimulated the formation of macropinosomes in Raw 264.7 macrophages [23], we measured macropinocytosis in MDA-MB-231 cells transfected with CA-Rac1.…”

“…Alternatively, studies by Swanson and colleagues have proposed that macropinocytotic cups can serve as signaling platforms that enhance GPCR signaling. In bone marrow-derived macrophages, inhibitors of macropinocytosis block stimulation of Akt by the CXCR4 receptor, and both PIP 3 and phosphory-lated Akt can be detected in macropinocytotic cups that form in CXCL12-stimulated bone marrow-derived macrophages [22]. In MDA-MB-231 cells expressing wild-type p110β, the expression of CA-Rac1 causes a large increase in the production of phase-bright vesicles, and a seven-fold stimulation of macropinocytosis.…”

“…Recent studies suggest that mature macropinosomes, enriched with PI(3,4,5)P 3 , can serve as efficient signaling platforms for GPCR-induced AKT phosphorylation and subsequent signal transduction 26,27 . Consistent with those studies, reduction of Rab10 expression, or treatment with the LRRK2 kinase inhibitor MLi2 to block pT73-Rab10 macropinosome maturation, both potently decreased CCL5-induction of AKT phosphorylation (Figure 9 A, B).…”

“…Macropinocytosis is a cellular process of solute endocytosis induced by growth factors, chemokines, and various other stimuli (Swanson, 2008, Egami et al, 2014, Buckley and King, 2017, Yoshida et al, 2018. In macrophages, stimulation with macrophage-colony stimulating factor (M-CSF) or the chemokine CXCL12 elicits membrane ruffles, which form cup-shaped structures that close into large endocytic vesicles called macropinosomes (Yoshida et al, 2009, Yoshida et al, 2015a, Pacitto et al, 2017. Macropinosomes either recycle to the plasma membrane or fuse with lysosomes.…”
