The naproxen bioavailability via percutaneous route is low, and several technologies have been used to overcome the problem. Although the permeation enhancer properties of natural essential oils have been reported, there is no study to show the effectiveness of Lavandula angustifolia essential oil on the percutaneous absorption of naproxen sodium from a topical gel to overcome poor percutaneous absorption of naproxen. To this end, the naproxen topical gel was formulated using Carbopol 940 (as a gelling agent), and several vehicles. The results showed a greater level of penetration into (222.19 ± 24.87 vs. 107.65 ± 6.38 µg/cm 2 ) and across (22.07 ± 4.42 vs. 13.14 ± 2.87µg/cm 2 ) the skin layers for the formulation containing essential oil in comparison with the naproxen gel ( P < 0.05) . A significant antinociceptive property was observed in naproxen topical gel containing 0.5% essential oil in both the first and late phase of the formalin test. The effect was observed in the late phase of the tail-flick test too. In conclusion, the study demonstrated that Lavandula angustifolia essential oil significantly enhanced the percutaneous absorption of naproxen and the analgesic effects.

Curcumin application as an anti‐cancer drug is faced with several impediments. This study has developed a platform that facilitates the sustained release of curcumin, improves loading efficiency, and anti‐cancer activity. Montmorillonite (MMT) nanoparticles were added to chitosan (CS)‐agarose (Aga) hydrogel and then loaded with curcumin (Cur) to prepare a curcumin‐loaded nanocomposite hydrogel. The loading capacity increased from 63% to 76% by adding MMT nanoparticles to a chitosan‐agarose hydrogel. Loading the fabricated nanocomposite in the nanoniosomal emulsion resulted in sustained release of curcumin under acidic conditions. Release kinetics analysis showed diffusion and erosion are the dominant release mechanisms, indicating non‐fickian (or anomalous) transport based on the Korsmeyer‐Peppas model. FTIR spectra confirmed that all nanocomposite components were present in the fabricated nanocomposite. Besides, XRD results corroborated the amorphous structure of the prepared nanocomposite. Zeta potential results corroborated the stability of the fabricated nanocarrier. Cytotoxicity of the prepared CS‐Aga‐MMT‐Cur on MCF‐7 cells was comparable with that of curcumin‐treated cells (p < 0.001). Moreover, the percentage of apoptotic cells increased due to the enhanced release profile resulting from the addition of MMT to the hydrogel and the incorporation of the fabricated nanocomposite into the nanoniosomal emulsion. To recapitulate, the current delivery platform improved loading, sustained release, and curcumin anti‐cancer effect. Hence, this platform could be a potential candidate to mitigate cancer therapy restrictions with curcumin.

Curcumin, is a diverse natural pharmacological agent involved in various signal transduction mechanisms. Therapeutically, this potent molecule facing different challenges and issues related to low bioavailability due to its poor aqueous solubility, less permeability, faster elimination and clearance. Experts in synthetic chemistry and pharmaceuticals are continuously sparing their efforts to overcome these pharmacokinetic challenges by using different structural modification strategies and developing novel drug delivery systems. In this mini‐review article, we are focusing on different possible routes like conjugation with biomolecules, natural polymers, synthetic polymers, natural products, metal conjugates and co‐administration with natural metabolic inhibitors and preparation of modified formulations such as micelles, microemulsions, liposomes, complexes with phospholipids, micro and nanoemulsions, solid lipid nanoparticles, nano lipid carriers, biopolymer nanoparticles and microgels to improve the pharmacokinetic properties of the curcumin without altering its pharmacodynamics activity. Hope this review helps to understand the problems associated with curcumin and different strategies to improve the pharmacokinetic profile of the curcumin.
