Abstract: Dense cell cultivation of the recombinant cell line BHK 21 pSVIL2 was performed in a fluidized bed bioreactor system containing porous borosilicate glass carriers. Experiments were carried out with different medium formulations for a period of 48 days. Due to an effective immobilization of the cells in the reactor, continuous operation was easy to perform. Maximal cell densities and product yields could be maintained, even when protein-free medium was perfused exceeding 2 reactor volumes per day. Final cell de… Show more

“…Earlier studies into the potential of fluidized-bed reactors for the production of biologics focused on the use of porous glass beads. These studies conducted by Kratje et al [77,79] examined the potential of fluidized-bed reactors as production systems using hybridomas suspension cells and anchorage-dependent BHK cells for the production of IL-2 (interleukin-2). In these studies, cell densities of 3.8 × 10 8 cells/ml intrasphere volume were reached, while the IL-2 production rate was 0.75 mg · l −1 · day −1 .…”

“…The immobilized cell densities and hFasL productivities can be maintained at high levels over a long period of time by continuous operation. The last cultivation runs under capsules recycling resulted in virtually no increase in cell densities, and this is due to the effect of intracellular metabolites inhibition as the nutrients were renewed . In terms of production levels, this study obtained a ∼20% higher productivity of hFasL than that reported by Lu et al in their work which produced hFasL by immobilized D. discoideum on porous matrices consisting of broken pumice or ceramic catalyst carrier.…”

“…A model process using a BHK‐21 cell line for the production of recombinant human interleukin‐2 in an airlift bioreactor system was established, applying a batch cultivation strategy. The BHK‐21pSVIL2 cell line was used before in several investigations on the production of recombinant human interleukin‐2 using different process conditions, including the application of protein‐free media (Lucki‐Lange and Wagner, 1991; Kratje and Wagner, 1992; Kratje et al, 1994). Prior to the experiments described here, the producer cell line used was adapted to protein‐free SMIF7 medium for optimal growth and productivity in single cell suspension cultures without forming spheroids, as shown in Figure 1, using a 2.5‐L membrane‐aerated bioreactor system (MSR2.5, Lehmann et al, 1987; Wagner and Lehmann, 1988).…”

“…The generation doubling time was reported to be 72 h in MSR2.5 using a 1:1 mixture of DMEM and Ham's F12 protein‐free medium (Ryll et al, 1990). An even higher generation doubling time of 115.1 h was observed for BHK‐21pSVIL2 in a fluidized bed bioreactor system using porous borosilicate glass beads as immobilization material for cell entrapment, and the same protein‐free medium (Kratje and Wagner, 1992; Kratje et al, 1994). However, 15.6% more LDH was released by the cells in the ALR30−SMIF7p system.…”
