Crystallization and Preliminary X-ray Diffraction Studies of New Crystal Forms ofEscherichia coliPIIComplexed with Various Ligands

Abstract: New crystals of the signal-transducing protein P(II) have been obtained in the presence of a number of different effector ligands. Various crystal forms are observed depending on the nature of the ligand(s). Co-crystallization with 2-ketoglutarate, glutamate and pyrophosphate produces hexagonal crystals similar to the wild type, ATP yields cubic crystals and ATP in conjunction with 2-ketoglutarate or glutamate yields orthorhombic crystal forms. All of the above crystals have been characterized by X-ray diffrac… Show more

“…The form II crystals were obtained at 277 K with the protein concentrated to 18–25 mg·mL −1 and stored in buffer A with 20 m m spermidine and a fourfold molar excess of ATP and α‐KG. The form II crystals were prepared from a solution that contained sodium acetate, poly(ethylene glycol) 3400 and Hepes pH 7.5 as described previously [18]. The space group was I222.…”

“…Data for both crystal forms were processed using the hkl program [19] and the results are shown in Table 1. Data for the form II crystals were collected at 100 K as described previously [18]. The form III crystals were mounted in glass capillaries and data were collected at 293 K. The first data set (form IIIa) was collected with a crystal that had been transferred to the reservoir buffer of the crystallization experiment while the second (form IIIb) was collected with a crystal soaked in 10 m m ATP, 10 m m CaCl 2 , 10% isopropanol and 100 m m sodium acetate pH 4.7.…”

“…A second, independent test of the need for uridylylation of GlnK to relieve NifL inhibition investigated the effects of a mutant form of GlnK in which the presumed site of uridylylation, Y51 (1,11,13,23,36,50), was altered. Western blot analysis ( Fig.…”
