Abstract: Besides size and polymorphic form, crystal shape takes a central role in engineering advanced solid materials for the pharmaceutical and chemical industries. This work demonstrates how multiple cycles of growth and dissolution can manipulate the habit of an acetylsalicylic acid crystal population. Considerable changes of the crystal habit could be achieved within minutes due to rapid cycling, i.e., up to 25 cycles within <10 min. The required fast heating and cooling rates were facilitated using a tubular reac… Show more

“…Tubular crystallizers such as the one presented here are characterized by a high surface-to-volume ratio, facilitating fast heat transfer and thus equilibration with the surrounding temperature is achieved after very short times. During the short residence times realized in our setup for each temperature step, no equilibration of the actual concentration with the compound's solubility can occur, as described for similar setups in our earlier publications [18,19]. Still, the larger the temperature swing is, the more crystal mass is dissolved and regrown per cycle due to accelerated rates of crystal growth and dissolution at higher super-and undersaturations, respectively.…”

“…A setup based on a continuously operated tubular crystallizer [15][16][17] and similar to our recently described tubular temperature cyclers [18,19] has been adapted to facilitate continuous deracemization. Polysiloxane tubing (d in = 2mm, d out = 4mm) with a total length of 50 m was coiled up into loops of 4 m each and immersed into two water baths to pass these alternately.…”

“…With slug flow, the homogenous flow conditions do not have to be fulfilled for stable operation. Investigations with slug flow were made regarding protein crystallization [59,60], alternating temperature profiles [45,61], product quality [44,62], nucleation without ultrasound [63], and nucleation with ultrasound [23,64].…”

“…[ 1 ] For example, a regular crystal habit and avoiding needles and flakes are beneficial for downstream powder handling. [ 2,3 ] Microparticle production with narrow size distribution is an essential step to provide acceptable dissolution properties for the formulation of poorly water‐soluble API. [ 4 ] Since crystallization is a core technology that impacts the quality of API in pharmaceutical industry, several precise control and intensified strategies have been reported.…”
