Creation of a watermelon haploid inducer line via ClDMP3-mediated single fertilization of the central cell

Abstract: Creating doubled haploids is one of the most efficient breeding methods in modern agriculture. Radiated pollen grains could induce haploids in cucurbit crops, which leads to a hypothesis that radiated pollen results in the preferential fertilization of the central cell over the egg cell was the reason of haploid formation. Disruption of DMP gene caused single fertilization of the central cell, which further induced haploids formation in plants. In the present study, a detailed method of creating watermelon hap… Show more

“…However, these agronomic traits need continuous breeding process to be fixed. It would be useful to develop a haploid inducer line to shorten the stabilization process of these lcltp3b homozygous lines ( Gilles et al., 2017 ; Chen et al., 2023 ). The lcltp3b homozygous lines are the results of different genetic combinations of M11 and other cultivars; thus, they can be directly used as breeding materials in searching for favorable agronomic traits in in the allergen-less background.…”

“…Loss of their function leads to either single fertilization of the egg cell or the central cell. Interestingly, in rare instances, central cell single fertilization can trigger parthenogenesis in the egg cell, resulting in the generation of haploid offspring (Zhong et al, 2020; Li et al, 2022; Wang et al, 2022a; Chen et al, 2023; Tian et al, 2023). Sexual reproduction in plants is a highly regulated and coordinated process influenced by various factors.…”

“…Rarely, following the defective single fertilization event, the egg cell has the potential to develop into an embryo, resulting in the production of maternal haploids at a low frequency (Zhong et al, 2020). Successful induction of haploid offspring in various species, such as tomato, watermelon, Medicago truncatula , and Brassica napus , have been achieved using dmp mutants as male parents (Li et al, 2022; Zhong et al, 2022; Wang et al, 2022a; Chen et al, 2023; Tian et al, 2023). However, the haploid induction ratio (HIR) remains relatively low, approximately 1%–2%.…”

“…DMP genes are conserved in dicots and have been shown to be involved in gamete fusion during double fertilization [ 10 ]. The Cas9/gRNA-mediated successful creation of HI lines by knockdown of DMP homologs in a variety of plants, including maize [ 11 ], tomato [ 12 ], potato [ 13 ], watermelon [ 14 ], Brassica napus , and Nicotiana tabacum [ 15 ], suggests their potential roles in the haploid induction process.…”
