Status epilepticus (SE) is a frequent medical emergency that can lead to a variety of neurological disorders, including cognitive impairment and abnormal neurogenesis. The aim of the presented study was the in vitro evaluation of potential neuroprotective properties of a new pyrrolidine-2,5-dione derivatives compound C11, as well as the in vivo assessment of the impact on the neurogenesis and cognitive functions of C11 and levetiracetam (LEV) after pilocarpine (PILO)-induced SE in mice. The in vitro results indicated a protective effect of C11 (500, 1000, and 2500 ng/mL) on astrocytes under trophic stress conditions in the MTT (3-[4,5-dimethylthiazole-2-yl]-2,5-diphenyltetrazolium bromide) test. The results obtained from the in vivo studies, where mice 72 h after PILO SE were treated with C11 (20 mg/kg) and LEV (10 mg/kg), indicated markedly beneficial effects of C11 on the improvement of the neurogenesis compared to the PILO control and PILO LEV mice. Moreover, this beneficial effect was reflected in the Morris Water Maze test evaluating the cognitive functions in mice. The in vitro confirmed protective effect of C11 on astrocytes, as well as the in vivo demonstrated beneficial impact on neurogenesis and cognitive functions, strongly indicate the need for further advanced molecular research on this compound to determine the exact neuroprotective mechanism of action of C11.

The history of seizures is amalgamated with the history of human existence, the first reports on epilepsy can be traced back to the Assyrian texts almost 2000B.C[1] A seizure is an extreme rush of electrical activity in the brain that can cause a variety of symptoms, depending on which parts of the brain are involved. Juvenile Seizuresare a common disorder that can be triggered or motiveless which are called provoked and unprovoked seizures respectively. Seizures, that are triggered by fever in young children or by severe hypoglycemia, are not considered to be forms of epilepsy. Unprovoked seizures have no clear cause but can be related to genetics or brain injury. When a child has two or more unprovoked seizures repeatedly, then it can be established as Epilepsy. Seizures not only affect the health of a child but also affect the entire family and are associated with increased parental stress with a decrease in quality of life (2) Epilepsy that begins within the first year of life often signifies increased morbidity. Although some epilepsyaffecting children less than oneyear of age can have favorable prognoses, others result in medically obstinate seizures, developmental delay/intellectual debility, and other neurologic impediments (3)(4). A classical description of Seizure disorders is available in various Siddha literature.The book yugi chinthamani-800, written by the great sage YUGIMUNI elaborately deals with both physical and mental illness.He described and classified various types of neurosis and functional psychosis.

: Epilepsy is a chronic neurological disorder, characterized by predisposition of unprovoked seizures affecting the neurobiological, psychological, cognitive, economic, and social well-being of patient. As per the 2019 report by World Health Organization, it affects nearly 80% of the population which comes from the middle to low-income countries. It has been suggested that 70% of such cases can be treated effectively if properly diagnosed. It is one of the most common neurological diseases affecting 50 million people globally. Most of the antiepileptic drugs used in clinical practice are only 60-80% effective in controlling the disease. These drugs suffer from serious drawbacks of non-selectivity and toxicity that limit their clinical usefulness. Hence, there is a need to search for the safe, potent, and effective anti-epileptic drugs. One of the emerging strategies to discover and develop selective and non-toxic anticonvulsant molecule focuses on design of non-nitrogen heterocyclic compounds (NNHC). Drugs such as valproic acid, gabapentin, viagabatrin, fluorofelbamate, tiagabine, progabide, pregabalin, gamma amino butyric acid (GABA) etc., do not contain a nitrogen heterocyclic ring but are as effective anticonvulsants as conventional heterocyclic nitrogen compounds. This review covers the various classes of NNHC which have been developed in the recent past as anticonvulsants along with their chemistry, percentage yield, structure activity relationship and biological activity. The most potent compound in each series has been identified for the comparative studies, for further structural modification and to improve the pharmacokinetic profile. Various optimized synthetic pathways and diverse functionalities other than nitrogen-containing rings discussed in the article may help medicinal chemists to design safe and effective anticonvulsant drugs in near future.
