Antimicrobial resistance is one of the greatest threats to human health. Alternatives to antimicrobials are needed to combat the rise of bacterial resistance. Essential oils (EOs) and their components are potential sources of new antimicrobials. The present study was conducted to evaluate the antibacterial and antifungal activities of two components of EOs. The antimicrobial mechanisms of eugenol and linalool were investigated against five bacterial strains and four Candida strains. Broth macrodilution method was used to compare the antibacterial and anticandidal activities of the two compounds. They exhibited antimicrobial activity against all tested strains. Germ tube formation by Candida albicans was investigated and it was found that it was completely inhibited at sub-MICs of eugenol while linalool showed minor activity compared to eugenol. Time kill kinetic studies indicated that eugenol was highly toxic to all bacterial and fungal strains within 2.5 h of exposure. Absorbance of intracellular constituents was measured at 260 nm. Only eugenol was highly effective toward lysis and cellular content leakage compared to control drugs. In addition, scanning electron microscopy (SEM) was used to charactize the effect of the two components on cell morphology and showed that both compounds induced cellular deformity of nearly all tested cells. Also, it was found that only eugenol inhibited the Beta-lactamase production and urease activity and it diminished bacterial motility of all tested bacterial strains. These results indicate that eugenol and linalool are effective antimicrobial agents and both antibacterial and antifungal activities of linalool were much weaker than that of eugenol.

Objective: The aim of this work was to assess the antifungal and post-antifungal effects of chlorhexidine, fluconazole, chitosan and its combinations on virulence factors of Candida albicans. Study Design: Ten isolated strains of Candida albicans obtained from 10 patients with oral candidiasis and a collection strain of C. albicans were treated with antifungal agents in different concentrations or combinations of them. Virulence factors analyzed were the cell surface hydrophobicity, the germinative tube development, the phospholipase activity and the post-antifungal effect of that exposure. Results: Virulence factors of the isolated strains obtained from patients together with the collection strain showed significant decreases with the different antifungal treatments, except for hydrophobicity and phospholipase activity. The development of germinative tube was the most sensitive factor to all the antifungal agents used. Untreated strains as well as the ones treated with antifungal agents showed a positive correlation among the virulence factors analyzed. No synergic effects arose from the combinations of the used drugs. Conclusions: C. albicans isolated strains from patients showed high phospholipase activity and germinative tube production, which corroborates their capacity to infect the oral mucosa and the high prevalence of species. As a whole, our results imply that short exposures to sub-inhibitory concentrations of the antifungal agents under analysis, isolated or combined, can modulate the way virulence factors get manifested, thus decreasing their pathogenicity.

Obecność w jamie ustnej ruchomych, płytowych uzupełnień protetycznych jest ważnym czynnikiem ryzyka powstawania stomatopatii protetycznych powikłanych zakażeniem grzybiczym. W powstawaniu i rozwoju zakażenia grzybiczego ważną rolę odgrywają enzymy hydrolityczne. Cel pracy. Przeprowadzono identyfikację gatunkową szczepów grzybów drożdżopodobnych u użytkowników pły-towych, ruchomych uzupełnień protetycznych, u których stwierdzono stomatopatię protetyczną oraz oceniono aktywności enzymów hydrolitycznych w uzyskanych izolatach i w bezpośrednich próbkach moczu. Materiał i metody. Badanie przeprowadzono wśród 20 pacjentów, u których stwierdzono stomatopatię protetyczną, bez cech zaburzonej immunokompetencji układu odpornościowego. Pobrano 40 próbek materiału biologicznego, na które składało się 20 wymazów z podniebienia oraz 20 próbek moczu. W badanych próbkach oznaczano gatunki grzybów drożdżopodobnych oraz wykonywano oznaczenie aktywności enzymów hydrolitycznych. Wyniki. Na podstawie hodowli z wymazów z podniebienia uzyskano 16 szczepów Candida albicans i 5 szczepów Candida tropicalis. Badanie aktywności hydrolaz w uzyskanych izolatach Candida albicans pozwoliło zakwalifikować badane grzyby do: biotypu A w 8 przypadkach, biotypu C3 -w 6, biotypu B1 -w 2 oraz C2 -w 1. W próbkach moczu nie wykazano wzrostu kolonii grzybów drożdżopodobnych, stwierdzono natomiast aktywność enzymów hydrolitycznych. Wnioski. Wśród użytkowników płytowych protez ruchomych, u których stwierdzono stomatopatię protetyczną dominującym gatunkiem w uzyskanych izolatach była Candida albicans. W stomatopatiach protetycznych powikłanych zakażeniem grzybiczym zaobserwowano uwalnianie enzymów hydrolitycznych do moczu bez cech kandydurii. Wraz ze zwiększającym się zaawansowaniem stadiów stomatopatii protetycznej zwiększała się także wirulentność izolowanych szczepów grzybiczych (Dent. Med.
