Cardiovascular disease (CVD) is one of the most important causes of human death, but no attention has been paid to cardiovascular health genes related to healthy longevity. Therefore, we developed a cohort study to explore such genes in healthy, long-lived Chinese subjects. A total of 13275 healthy elderly people were enrolled, including 5107 healthy long-lived individuals and 8168 age-matched control individuals with low CVD risk. Using a combination of whole-exome sequencing (WES) and genome-wide association studies (GWAS), we identified 2 genetic variants (TFPI rs7586970 T, p=0.013, OR=1.100. ADAMTS7 rs3825807 A, p=0.017, OR=1.198) associated with healthy lipid metabolism and longevity. Furthermore, we showed that an interaction among TFPI rs7586970, ADAMTS7 rs3825807 and APOE ɛ3 maintained normal blood lipid levels in centenarians by stratified analysis of CVD risk factors. Finally, through biological function analysis, we revealed clues regarding the mechanism of factor related to cardiovascular health (FCH) such as lipids and longevity. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis indicated that the two variants above may be associated with longevity via FCH lipid metabolism pathways. From a meta-analysis of venous thrombosis patients, we unexpectedly found that rs7586970 T is associated with both longevity and protection against vascular disease.

Adenosine (A) to inosine (I) RNA editing is the most prevalent RNA editing mechanism in humans and play critical roles in tumorigenesis. However, the effects of radiation on RNA editing and the mechanisms of radiation-induced cancer were poorly understood. Here, we analyzed human bronchial epithelial BEP2D cells and radiation-induced malignantly transformed cells with next generation sequencing. By performing an integrated analysis of A-to-I RNA editing, we found that genome-encoded single-nucleotide polymorphisms (SNPs) might induce the downregulation of ADAR2 enzymes, and further caused the abnormal occurrence of RNA editing in malignantly transformed cells. These editing events were significantly enriched in differentially expressed genes between normal cells and cancer cells. In addition, oncogenes CTNNB1 and FN1 were highly edited and significantly overexpressed in cancer cells, thus may be responsible for the lung cancer progression. Our work provides a systematic analysis of RNA editing from lung tumor specimens with high-throughput RNA sequencing and DNA sequencing. Moreover, these results demonstrate further evidence for RNA editing as an important tumorigenesis mechanism.

Background: The human microbiome is a mixed group of microorganisms and has been verified the relationship between gastrointestinal microbiota and blood pressure. The intestinal microbiota of chronic kidney disease (CKD) patients in the distribution of bacterial species is different from the flora of people with no CKD. The purpose of this research is to study on the different intestinal microbiota of hypertensive patients with and without nephropathy and to find possible bio-markers to diagnose hypertensive nephropathy, as well as one of the basis for hypertensive nephropathy or CKD clinical intervention treatment. Methods: The study subjects were divided into three groups, normal control, simple hypertension and hypertensive nephropathy. Sequencing, bioinformatics and statistical analysis were performed on the 16S rRNA gene of the flora of the subjects' stool samples. Results: This research shows the difference of intestinal flora in hypertension patients with and without nephropathy; it investigates the relationship of the differences in the intestinal microbiota with bile-acid metabolism; it also explores bile-acid metabolism mechanism of intestinal microbiota differences of hypertensive with or without nephropathy. Conclusions: The different gastrointestinal microbiota is related to bile-acid metabolism in hypertensive patients, and can be one of the factors causing CKD. Trial registration: This article does not report the results of a health care intervention on human participants.
