Abstract: A unique feature of human alveolar macrophages is their prolonged survival in the face of a stressful environment. We have shown previously that the ERK MAPK is constitutively active in these cells and is important in prolonging cell survival. This study examines the role of the ERK pathway in maintaining mitochondrial energy production. The data demonstrate that ATP levels in alveolar macrophages depend on intact mitochondria and optimal functioning of the electron transport chain. Significant levels of MEK a… Show more

“…For example, ERK has been reported to be an important macrophage survival factor in some (35)(36)(37)(38)(39)(40) but not all (30,41,42) studies, and we detected decreased ERK phosphorylation in response to LPS or thapsigargin in BMDMs from both WT and BCAP-deficient mice, as well as a trend for enhanced ERK phosphorylation in unchallenged BMDMs from BCAP-deficient mice (Fig. 4).…”

“…32,34,35 This study provides evidence that LX might affect these pathways. Incubation of macrophages with staurosporine induces a rapid activation of JNK and p38 MAPKs, but not of ERK, a classic survival pathway.…”

“…135,136 Additional cells in which ERK1/2 were identified in the mitochondria include murine cardiac myocytes, where the kinases are associated with enhanced Bad phosphorylation and cardioprotection, 137 and human alveolar macrophages, where ERK1/2 maintain mitochondrial membrane potential and M Monographs ATP production. 138 Finally, detailed immunoelectron microscopy studies have established the presence of phosphorylated ERK1/2 in the outer membrane and intermembrane space of brain mitochondria, 139,140 suggesting their role in multiple mitochondria functions.…”
