Abstract: As a distinct type of head and neck cancer, non-keratinizing nasopharyngeal carcinoma (NPC) is closely associated with EBV infection and massive lymphoid infiltration. The unique histological features suggest that local inflammation plays an important role in NPC tumourigenesis. We comprehensively characterized NF-κB signalling, a key inflammatory pathway which might contribute to the tumourigenesis of this EBV-associated cancer. By EMSA, western blotting, and immunohistochemical staining, constitutive activat… Show more

“…Protein encoded by Helicobacter pylori inhibited the p53 pathway by increasing the ubiquitination of p53 to promote cell migration in gastric cancer (Wei et al ., 2010). The nuclear factor‐κB signaling pathway was activated by Epstein–Barr virus in nasopharyngeal carcinoma and contributed to tumor metastasis (Chung et al ., 2013). In the present study, HBx directly interacted with AIM2 to enhance its ubiquitination, subsequently resulting in a rapid degradation of AIM2 protein.…”

“…It is also possible that the commonly observed infiltration of intratumoral lymphocytes in NPC may limit the ability to detect some somatic mutations that are present in only a small fraction of the population of the cancer cells. NF-κB signaling has been extensively implicated in NPC, with constitutive activation detected in NPC cell lines and primary tumors (26). Several mechanisms have been proposed for NF-κB signaling activation in NPC.…”

“…Glut-1 inhibitors with potential applications in cancer treatment have recently been developed. STF-31, used in this study, is one of the promising Glut-1 inhibitors which have been shown to selectively kill renal carcinoma cells (25,43). Interestingly, there are reports showing that high glucose levels also enhance virus infection of host cells, supporting viral replication and maintenance of latency infection (44)(45)(46).…”

“…IKK activation is the upstream event of NF-B activation. Constitutive NF-B activation is common in EBV-associated NPC and is involved in NPC pathogenesis (43). Activation of NF-B is a major signaling pathway of the EBV-encoded LMP1.…”
