Abstract: We aimed to identify the functional characteristics of the corpus luteum (CL) by color Doppler ultrasonography and changes in interferon-stimulated gene (ISG) expression in peripheral blood mononuclear cells (PBMCs) during early pregnancy in beef cows. We then aimed to use these features to establish earlier pregnancy diagnosis methods. In experiment 1, the CL size and blood flow were accessed by Doppler ultrasonography, and the PBMCs were isolated on Days 8, 12, 15, 18, 20, 22, 25, 30, 45, and 60 post-timed a… Show more

“…Significant differences in mRNA abundance were found between pregnant and non-pregnant ewes with an up-regulation of MX1 gene expression in pregnant animals as previously described in sheep (Yankey et al, 2001) and cattle (Gifford et al, 2007;Pugliesi et al, 2014). Furthermore, if CXCL10 and STAT1 (Gray et al, 2006) mRNA levels are known to be up-regulated by pregnancy in the ovine endometrium, we observed that they were also up-regulated in PBMCs during early pregnancy.…”

“…Therefore real-time PCR analyses of ISG mRNA expression were run with different populations of circulating cells. Expression of ISG (ISG15, MX1, MX2) has been already described in PBMC (Shirasuna et al, 2012;Kizaki et al, 2013;Pugliesi et al, 2014), and in leucocytes (Yankey et al, 2001;Gifford et al, 2007;Green et al, 2010a). Kinetics of ISG expression level during peri-implantation was shown to be similar in PBMC and PMN (Shirasuna et al, 2012).…”

“…In another study in pregnant ewes, the MX1 transcript levels was significantly upturned on days 15, remained consistent till days 23 and then declined on days 25-30 28 . Furthermore, the MX1 expression was also noted to be elevated on days 15 in PBMCs of pregnant ewe 32 and cow 33 . In our recent study, the MX1 gene registered greatest expression on days 14, then gradually down-regulated up to days 20 and again increased on days 22 in pregnant buffaloes 15 .…”
