“…For example, Tfh-like cells, traditionally thought to be a relatively uniform subset of cells, can be sub-classified into 15 distinct subclusters when analyzed by CyTOF (Wong et al, 2015). While CyTOF was used recently to compare CD4+ T-cell subsets in uninfected and ART-suppressed HIV-infected subjects (Corneau et al, 2017), it has not been used to characterize cellular susceptibility to HIV infection.…”

“…In a study from our lab, we measured phospho-signaling differences in uninfected and latent HIV-infected T cells and used PLS-DA to define signatures that predicted infection status and then identified changes in stress kinase signaling as a novel target for preferential infected-cell killing [40]. Comparing total immune cell composition in healthy and infected donors, SPADE and t-SNE analyses performed on 26-parameter mass cytometry data demonstrated that infected patients had a higher fraction of highly differentiated effector memory cells following HIV infection and ART treatment in comparison to healthy donors [41]. Overall, systems immunology studies of virus-host immune interactions have generated new hypotheses about mechanisms of viral pathogenesis and potential strategies to combat viral infection.…”

“…MC allows for deep profiling of the heterogeneity inherent in biological samples to uncover rare cell populations which may have profound influences in health and disease. Recent examples enabled by MC include the discovery that the brain's reward system affects immunity , identification of HIV‐containing CD4 + T cell reservoirs , elucidation of the role non‐IgE‐mediated immune responses play in food allergies , exploration of the biological basis for lead neurotoxicity , and discovery that coordinated peripheral immunity is required for tumor eradication .…”
