Compound danshen dripping pills modulate the perturbed energy metabolism in a rat model of acute myocardial ischemia

Abstract: The continuous administration of compound danshen dripping pills (CDDP) showed good efficacy in relieving myocardial ischemia clinically. To probe the underlying mechanism, metabolic features were evaluated in a rat model of acute myocardial ischemia induced by isoproterenol (ISO) and administrated with CDDP using a metabolomics platform. Our data revealed that the ISO-induced animal model showed obvious myocardial injury, decreased energy production, and a marked change in metabolomic patterns in plasma and h… Show more

“…Isoproterenol (ISO) is a strong β-adrenergic agonist, which can increase myocardial oxygen consumption through speeding up heart rate and enhancing myocardial contractility, resulting in cardiac overload, myocardial microcirculation disturbance, coronary artery spasm, myocardial infarction, myocardial necrosis. As it is widely accepted that Isoproterenol (ISO) injection can readily induce acute myocardial ischemia injury in rats ( Shi et al, 2013 ; Guo et al, 2016 ; Dhivya et al, 2017 ).…”

“…The mechanism of ISO-induced myocardial ischemia is due to the contracted coronary artery, strengthened cardiac contractility, increased myocardial oxygen consumption, and finally resulted in myocardial infarction in short time [30]. Mice injected with ISO led to acute myocardial ischemia which increased the demand of oxygen supply, and once the oxygen was in short supply, the body could not afford time to complete the compensatory response and eventually resulted in death of heart energy shortage.…”

“…Finally, the solution was vortex-mixed again for 30 s after adding the external standard, methyl myristate in heptane (30 μg/mL), to each GC vial for the GC-mass spectrometry (MS) analysis. GC/MS analysis GC/MS analysis was performed, as previously described [24]. In brief, chromatographic separation of the analytes was achieved with a Shimadzu GCMSQP2010 (Shimadzu Corp., Tokyo, Japan) equipped with a RTx-5MS column (30 mm × 0.25 mm i.d.…”
