Competitive Chemiluminescent Immunoassay for Estradiol Using anN-Functionalized Acridinium Ester

Abstract: We have compared three competitive chemiluminescent immunoassays (CLIA) for estradiol (E2) using an N-functionalized acridinium ester (AE). The assays were a standard competitive assay using immobilized antibody and directly labeled antigen (type A), an immobilized antibody and indirectly labeled antigen (type B), and an immobilized antigen and labeled antibody (type C). In an antibody-immobilized system, the assay using both AE- and E2-labeled thyroglobulin as a tracer (type B) was more sensitive than that us… Show more

Exaggerated anticipatory anxiety is common in social anxiety disorder (SAD). Neuroimaging studies have revealed altered neural activity in response to social stimuli in SAD, but fewer studies have examined neural activity during anticipation of feared social stimuli in SAD. The current study examined the time course and magnitude of activity in threat processing brain regions during speech anticipation in socially anxious individuals and healthy controls (HC). Method Participants (SAD n = 58; HC n = 16) underwent functional magnetic resonance imaging (fMRI) during which they completed a 90s control anticipation task and 90s speech anticipation task.

“…Acridine ester is a high effective label in chemiluminescence immunoassay (CLIA) [4], and is conjugated with amino, carboxyl or phosphate groups which are from biomacromolecule such as antigen, antibody and DNA for development of CLIA [5][6][7][8]. In 1983, Ian Weeks et al [9] improved CLIA using acridine ester as a label which has less background and better sensitivity [10][11][12][13]. The sensitivity, repeatability and accuracy of CLIA are preferable to ELISA [14].…”

“…9,10 Acridinium ester has been usually used as the CL label in immunoassays due to its extremely high sensitivity and selectivity. [11][12][13][14][15] Since CL was firstly used in immunoassays, 12 a variety of configurations have been introduced in assays of clinical importance. Flow injection analysis (FIA) used in association with CL immunoassays has proved to offer advantages of faster speed and better reproducibility.…”
