Abstract: Introduction: Clinical doses of anaesthetic agents were administered to rabbits and effects on the brain, heart, and liver were investigated biochemically and histopathologically. Material and Methods: The rabbits were randomly divided into three main groups (16 rabbits each) and each group into study (n = 8) and control (n = 8) groups. All study group rabbits received 3 mg/kg of midazolam (M) intramuscularly. Group 1.1 (M) received nothing further, group 2.1 (MK) also received 25 mg/kg of ketamine, and group … Show more

“…In the present study, moderate hypoxia, hypercapnia and acidosis were found in DXK, whereas mild hypoxia, hypercapnia and acidosis occurred in MDK. In agreement with findings of other researchers except Nishida et al (26), as a result of suppression of the respiratory system by the anaesthetics, these changes in both groups were noticeable especially within the first 15 min (2,7,10,18,20). This change may have been affected by the fact that the pO 2 value at the baseline in DXK was significantly lower (p < 0.05), and O 2 supplementation was provided in both groups at the end of the 120 min.…”

“…It has been emphasised that α2 agonist-ketamine combinations lead to hypoxia, hypercapnia and acidosis in rabbits, and that, when using these combinations, O 2 supplementation is necessary (7,16,17,20,27,29). It was reported that, although dexmedetomidine depressed ventilation, it did not lead to deep hypoxemia or hypercapnia (26), while the midazolam-ketamine combination may have caused moderate respiratory acidosis by leading to changes in blood gasses, especially at the 30 th and 60 th minutes (2). In the present study, moderate hypoxia, hypercapnia and acidosis were found in DXK, whereas mild hypoxia, hypercapnia and acidosis occurred in MDK.…”

“…While V T was similar between awake and asleep stages, the RR was lower during sleep. A possible explanation for these observations is anxiety-related hyperpnea of rabbits during the awake stage ( Schroeder and Smith, 2011 ; Atalan et al, 2019 ) and its cessation during sleep. Furthermore, a lower metabolic rate and chemosensitivity to both oxygen ( Douglas et al, 1982b ) and carbon dioxide during sleep ( Douglas et al, 1982c ) might explain a diminished RR and MV.…”

“…In rabbits, various anesthetics have differing effects on biochemical indicators and even organ histology 7 , and variations in the tolerance of individual rabbits to anesthetics will result in different anesthetic effects among experimental rabbits 23 . Matchett and Wood 24 also found that anesthesia affected heart rate variability, which further limits the use of anesthetics in animal experiments.…”

“…However, anesthesia affects the respiration, blood pressure, and heart rate of the experimental animals 5 , 6 . It can lead to apoptosis of rabbit tissue cells 7 , making it particularly difficult for histopathological studies to control for variables. The differences in the tolerance levels of different individual rabbits to anesthesia, awakening during the extended examination procedure, and postural changes in multiple scans may affect the imaging results.…”
