Comparison of ISSR and SSR markers for analysis of genetic diversity in the seagrass Posidonia oceanica

Abstract: Posidonia oceanica is an endemic seagrass species in the Mediterranean Sea. Due to its ecological importance, it has been the subject of several genetic studies aimed at increasing general knowledge of the system and helping to define appropriate management strategies. We compared the efficiency of 2 different classes of molecular markers, 'inter simple sequence repeats' (ISSRs) and 'simple sequence repeats' (SSRs) or 'microsatellites', in population genetic analysis of P. oceanica. To do this, we analyzed mea… Show more

“…Calculated P sex probabilities were all lower than 0.01, which is the level that was used to reject the null hypothesis that the ramets belong to individuals derived from distinct sexual events (Serra et al . ). We used MLGs for all following statistics, but the number of MLLs is also reported.…”

“…However, as somatic mutations and scoring errors could lead to an underestimation of the number of clones, the data set of MLGs was further investigated by removing one locus at a time to identify MLGs that are distinct at one locus, termed MLLs and re-calculating P sex . Calculated P sex probabilities were all lower than 0.01, which is the level that was used to reject the null hypothesis that the ramets belong to individuals derived from distinct sexual events (Serra et al 2007). We used MLGs for all following statistics, but the number of MLLs is also reported.…”

“…By molecular techniques, many authors estimated seagrass genetic variability (Reusch 2001;Micheli, Paganin, Peirano, Caye, et al 2005;Diaz-Almela et al 2007;Procaccini et al 2007;Schwartz et al 2007;Serra et al 2007) and assessed the impact of disturbance on gene flow using random amplified polymorphic DNA (RAPD) technique (Waycott 1995;Cheng et al 1997;Amzallag 1999;Alberto et al 2001;Jover et al 2003;Jones et al 2008). For all these reasons, we used them to verify the probability of an external genetic input by comparing the genetic traits of fruits collected in one P. oceanica meadow with those of fruits stranded in front of the meadow and downcurrent.…”
