Comparison of aqueous, polyethylene glycol-aqueous and ethanolic propolis extracts: antioxidant and mitochondria modulating properties

Abstract: BackgroundPropolis is multicomponent substance collected by honeybees from various plants. It is known for numerous biological effects and is commonly used as ethanolic extract because most of active substances of propolis are ethanol-soluble. However, water-based propolis extracts could be applied more safely, as this solvent is more biocompatible. On the other hand, water extracts has significantly smaller range and quantity of active compounds. The extraction power of water could be enhanced by adding co-so… Show more

“…The oxygen uptake rates of mitochondria in C6 cells were recorded using high-resolution respirometer Oxygraph-2k (Oroboros Instruments, Innsbruck, Austria) as described in [19]. The measurement medium contained 0.5 mM EGTA, 3 mM MgCl 2 , 60 mM K-lactobionate, 20 mM Taurine, 10 mM KH 2 PO 4 , 20 mM HEPES, and 110 mM sucrose (pH 7.1 at 37 °C).…”

“…Phenolic acids and aldehydes make up 40–42% of all extracted and identified compounds in aqueous (AqEP) and polyethylene glycol–aqueous (Pg-AqEP) propolis extract, and only 16% in ethanolic propolis extract (EEP) [ 23 ]. All preparations revealed similar antioxidant activity in cell-free culture medium, but in a C6 astrocytoma cell culture, Pg-AqEP and EEP demonstrated better mitochondrial superoxide- and total intracellular ROS-decreasing properties [ 24 ]. In addition, Pg-AqEP had a concentration-dependent, mitochondria-uncoupling effect, the property that might prevent irreversible opening of the mitochondrial permeability transition pore [ 25 ].…”

“…We used ethanolic extracts of propolis (EEP), which is the most common way of making propolis preparations. EEP is known for its highest content of biologically active propolis components, among which flavonoids comprise the main part [ 37 ]. Due to some limitations in medical use of strong ethanolic solution (70% alcohol in our case), we also prepared emulsions of EEP in water (1.4% alcohol).…”

“…Nevertheless, the recognized prerequisite for EZ formation is the presence of hydrophilic functional groups. Propolis is predominantly lipophilic in nature, but its constituents possess an abundance of hydrophilic -OH and -COOH functional groups [ 37 ], which may underlie the particle-exclusion capacity of the interfacial water next to propolis. The presence of those hydrophilic groups in all propolis samples, regardless of the source and variation of chemical composition, should assure the generic presence of EZ in the water next to propolis surfaces.…”
