Communication Impairment in Ultrasonic Vocal Repertoire during the Suckling Period of Cd157 Knockout Mice: Transient Improvement by Oxytocin

Abstract: Communication consists of social interaction, recognition, and information transmission. Communication ability is the most affected component in children with autism spectrum disorder (ASD). Recently, we reported that the CD157/BST1 gene is associated with ASD, and that CD157 knockout (Cd157 −/− ) mice display severe impairments in social behavior that are improved by oxytocin (OXT) treatment. Here, we sought to determine whether Cd157 −/− mice can be used as a suitable model for communication deficits by meas… Show more

“…We next examined the effect of oxytocin administration on USV emission of control and VPA pups on PND 11. We applied oxytocin by s.c. injection (10 ng/pup) as reported previously 19 and analysed the number of calls before and 30 minutes after the injection. Two‐way repeated measures ANOVA indicated a significant interaction between Time (before and 30 min after the injection) × [Treatment (control or VPA) and Drug (saline or oxytocin)] ( F 3,45 = 5.457, P = 0.003) (Table 5).…”

“…The average start frequency was analysed similarly to the average duration, except that each USV call was checked manually. The pattern of USV on PND 11 was manually classified using methods described previously 19 …”

“…Similar to Nrxn1α mice, Reeler mutant pups produced a narrowed repertoire of calls, specifically on PND 6 and 8, limiting their vocal repertoire to the two‐components , chevron and complex calls ; while Dab1 mutant pups showed a limited call pattern restricted to short and downward calls . A poor USV repertoire also has been detected in Cd157 KO pups from PND 3 until 10 compared with a rich repertoire in WT mice . These seemingly conflicting findings may simply indicate that the animal models reflect the diversity observed in patients across the different ASD, including the atypical vocalizations.…”

“…Importantly, an oxytocin analogue has long lasting effects on anxiety behavior in a CD157 knockout mouse (51) and communication impairment during the suckling period is restored by oxytocin in the CD157 knockout (52). CD157 and CD38 knockouts show decreased plasma oxytocin levels (53) suggesting a role for both, CD157 as well as CD38 in oxytocin (OT) release and thus as putative regulators of complex behaviors (53).…”
