Collection and Analysis of Respiratory Mucus from Subjects without Lung Disease

Abstract: We collected the respiratory mucus coating the endotracheal tubes used during short surgical procedures in 27 patients with no clinical evidence of respiratory disease. Twelve were male and 15 were female, and they ranged from 1 to 64 yr of age (mean, 28.7 yr). The viscoelastic properties, frog palate transport rate, and percent solid composition were in the normal range reported for both canine and human mucus collected using the bronchoscopy brush technique. There were no significant differences noted betwee… Show more

“…Healthy individuals produce much smaller amounts of airway secretions and do not expectorate mucus, but mucus samples from normal airways can be obtained following a standardized protocol of hypertonic saline exposure (270). Mucus samples can also be obtained by endotracheal suctioning of patients without airway diseases who are undergoing surgical procedures (231).…”

“…15 In our study the evaluation of macroscopic appearance of the secretions was always performed by 2 investigators. To reduce the subjectivity of those evaluations we used the scale described by Deneuville et al 9 Regarding the samples from the without-lung-disease subjects, the mucus analysis was difficult, because small amounts were collected, so for this group we applied the method described by Rubin et al, 8 in which mucus is collected from the endotracheal tube used during a short surgical procedure in without-lung-disease subjects. The characteristics of the mucus deposited inside and outside the tube was compared.…”

“…6 In the without-lung-disease subjects the samples were collected immediately after removal of the endotracheal tube. The secretion sample that was deposited inside the tube was removed with a swab, according to the criteria and procedures described by Rubin et al 8 Respiratory secretions were assessed as follows:…”
