Cognitive impairments caused by oxaliplatin and 5-fluorouracil chemotherapy are ameliorated by physical activity

Abstract: The combination 5FU/OX had a significant impact on cognition. However, rats treated with 5FU/OX that exercised post chemotherapy had improved cognition relative to non-exercising rats. This suggests that physical activity may prove useful in ameliorating the cognitive impairments induced by 5FU/OX.

“…By convention, we have induced bifurcation by introducing the animals to a novel wheel at the start of one of the scotophases [2], and therefore wheels were used in our protocol. Running wheels have been shown to change the organization of sleep [44], increase learning [45,46] and synaptic plasticity [47], and rescue induced learning deficits [48,49]. While it is unknown whether the wheels may have compensated for small decrements dependent on sleep or other variables, the fact that our protocol was sensitive to differences in circadian manipulation (Experiment 1) and phase (Experiment 2) in control animals indicates that the wheels did not create a general ceiling effect.…”

“…Further, the combination of oxaliplatin ? 5-fluorouracil produces deficits in novel object recognition 2 weeks following exposure [131] and methotrexate produces deficits in novel object recognition 4 weeks following a single exposure [129] indicating that these effects can persist.…”

“…Neonatal exposure to methotrexate can impair the development of conditioned emotional responding when assessed in young animals [144]. In adult animals, a single exposure to oxaliplatin and 5-fluorouracil results in deficits in contextual fear contitioning 4 weeks following exposure [131] and repeated exposure to cyclophosphamide and doxorubicin leads to impairments in contextual fear conditioning that persist for at least 1 week following the final exposure [145]. Additionally, repeated temozolomide exposure impairs acquisition of a classically conditioned eyeblink response [92].…”
