Circulating miRNAs and miRNA shuttles as biomarkers: Perspective trajectories of healthy and unhealthy aging

Abstract: Human aging is a lifelong process characterized by a continuous trade-off between pro-and anti-inflammatory responses, where the best-adapted and/or remodeled genetic/epigenetic profile may develop a longevity phenotype. Centenarians and their offspring represent such a phenotype and their comparison to patients with age-related diseases (ARDs) is expected to maximize the chance to unravel the genetic makeup that better associates with healthy aging trajectories. Seemingly, such comparison is expected to allow… Show more

“…MicroRNAs (miRNAs) are a type of small (~22-nt long), non-coding RNA molecules that regulate target gene expression by translational inhibition or mRNA destabilization through the combination with the 3'-untranslated region of target messenger RNAs. The loss of homeostasis in the miRNA/mRNA axis leads to a number of physiological and pathological events, such as inflammation, senescence and tumorigenesis (1,2).…”

“…Yang et al demonstrated that miR-598 suppressed the invasion and migration in NSCLC as a tumor suppressor through negatively regulate Derlin-1 (DERL1) and epithelial-mesenchymal transition (EMT) [10]. However, the roles of genes and miRNAs in NSCLCs are still not well understood [11].…”

“…Attaining this goal clearly requires more accurate and possibly more numerous biomarkers of biological aging capable of predicting functional capacity at a later age than chronological age. Since the variance of aging phenotypes widens over time, owing to different individual trajectories of aging (Ferrucci et al, 2020;Olivieri et al, 2017), immunological/biological age rather than chronological age appears to be a more sensitive approach also to assess individual susceptibility to the adverse outcomes of COVID-19 infection. Consequently, a panel of biomarkers of immunological/biological aging could help identify subjects at risk of adverse COVID-19 outcomes also among the younger population.…”
