Chronic arsenic exposure and urinary 8-Hydroxy-2′-deoxyguanosine in an arsenic-affected area in Inner Mongolia, China

Abstract: Objective: Recent studies have shown that generation of reactive oxidants during arsenic metabolism can play an important role in arsenic-induced injury. The purpose of this study was to examine the relationship between arsenic in drinking water and oxidative stress in humans by measuring 8-Hydroxy-2 0 -deoxyguanosine (8-OHdG). Methods: We performed a cross-sectional study in an arsenic-affected village in Hetao Plain, Inner Mongolia, China. A total of 134 of the 143 inhabitants (93.7%) of the village particip… Show more

“…The 8-OxodG levels detected were low compared to the other studies of As-exposed populations with similar exposure range [8][9][10][11]. It is important to note that the high 8-OxodG levels detected in other studies [8, [10][11][12][13][14][15] are partly due to the methods used; especially ELISA, the most commonly used method for 8oxodG determination, has been shown to overestimate 8-OxodG levels, whereas mass spectrometric techniques appear to be more robust [45].…”

“…As exposure and 8oxodG where they have employed similar LC/MS-MS methods higher 8oxodG levels were found: Fujino et al [9] reported mean 8-OxodG levels of 5.5 μg/g creatinine among subjects without As dermatosis and mean U-As levels of 500 μg/g creatinine; Hu et al [21] reported 8-OxodG levels of 5.6 μg/g creatinine among exposed semiconductor workers with U-As levels of 33 μg/g cre. Thus, the method used in the present study cannot explain the low concentrations of 8-OxodG in this population.…”

“…For this purpose, understanding the mechanisms of 8-OH-Gua generation is an important issue. As for arsenic compounds, many reports have indicated that they increased oxidative DNA damage in cultured cells (6,7). For example, hydroxyl radical-induced oxidative DNA damage was generated in arsenic trioxide-treated keratinocyes (10).…”

“…In particular, they are associated with the development of several kinds of tumors, including lung, skin, kidney, bladder and liver tumors (1)(2)(3)(4)(5). Recently, arsenic trioxide was shown to induce DNA damage, such as 8-hydroxyguanine (8-OH-Gua) and DNA strand breaks, in cultured human cells (6,7). It was also reported that the urinary 8-OH-dG level is increased with exposure to arsenic compounds (8,9).…”
