Chromosome organization in meiosis revealed by light microscope analysis of silver-stained cores

Abstract: Three species of grasshoppers have been analyzed by means of a modified silver impregnation technique that reveals the presence of a chromatid core that identifies chiasmata at first meiotic metaphase. In terms of the behaviour of the chromatid core most of the configurations observed at diplotene with orcein are easily recognized in metaphase I silver-stained bivalents. Some "hidden" configurations, as well as simple chromatin associations, that do not appear to represent chiasmata have also been detected. Th… Show more

“…For DAPI/DA staining, the slides were treated for 20 min with DAPI and 40 min with DA. Silver nitrate impregnation was performed as described by Rufas et al (1987). In brief, the slides were pre-treated with 2X SSC at 60°C for 10 min and stained with silver nitrate (1 g/mL) at 70°-80°C.…”

“…Slides used for C-banding, silver nitrate staining and FISH analysis were prepared in 45% acetic acid and coverslips were removed after freezing the preparations by immersing in liquid nitrogen for a few seconds. C-banding was performed according to the method described by Sumner (1972) and silver nitrate staining was conducted according to Rufas et al (1987).…”

“…• C. Triple combination of the fluorochromes CMA 3 /DA/DAPI and silver staining were performed using the protocols of Schweizer (1976) and Rufas et al (1987) respectively, and FISH followed the procedure of Moscone et al (1996), using a probe of 45S rDNA isolated from Arabidopsis thaliana. The images were captured in Leica DM LB microscope (Leica Microsystems, Wetzlar, Germany) and the figures were organized using the Corel Photo-Paint 12 software (Corel corporation 2003, Ottawa, Canada).…”
