Abstract: BackgroundThe cholesteryl ester transfer protein (CETP) polymorphism I405V has been suggested to be involved in longevity and susceptibility to cardiovascular diseases. An enhanced reverse cholesterol transport due to enhanced HDL levels has been hypothesized to be the underlying mechanism. However, clinical trials with HDL-enhancing drugs failed to show beneficial effects. Consequently, it has been postulated that genetic variations enhancing HDL levels are cardioprotective only if they also decrease LDL leve… Show more

“…We examined pegRNAs that install therapeutically relevant mutations associated with Alzheimer’s disease 44 , coronary heart disease 45 , 46 , type-2 diabetes 47 , innate immunity 48 , CDKL5 deficiency disorder 49 , lamin A deficiency 50 , and Rett syndrome 51 , 52 . We compared the outcomes of prime editing with pegRNAs and corresponding tevopreQ 1 epegRNAs with 8-nt pegLIT linkers in HEK293T cells ( Fig.…”

“…It has also been shown that the ‘VV’ genotype of the SNP rs5882 is associated with lower plasma CETP levels and increased HDL concentration [ 75 ]; however, baseline HDL data for participants with the ‘VV’ genotype was not available for all the studies because there were not enough participants with the ‘VV’ genotype in the two Iranian studies [ 65 , 66 ]. Also, in the two Inuit studies [ 16 , 71 ], baseline HDL data was not recorded separately for ‘VV’, ‘IV’ or ‘II’ genotype.…”

“…Thus, we failed to find an association between the HDL-cholesterol increasing genotypes of CETP to CVD. It must be noted that genetic mechanisms raising plasma HDL-cholesterol do not decrease the risk of myocardial infarction 25 , and only SNPs affecting LDL-cholesterol levels or both, LDL-cholesterol and HDL-cholesterol levels, influence CVD risk 26 .…”
