The mushroom, Sanghuang is widely used in Asian countries. This medicinal fungus produces diverse bioactive compounds and possesses a potent ability to degrade the wood of the mulberry tree. However, the genes, pathways, and mechanisms that are involved in the biosynthesis of the active compounds and wood degradation by Sanghuang mushroom are still unknown. Here, we report a 34.5 Mb genome-encoding 11,310 predicted genes-of this mushroom. About 16.88% (1909) of the predicted genes have been successfully classified as EuKaryotic Orthologous Groups, and approximately 27.23% (665) of these genes are involved in metabolism. Additionally, a total of 334 genes encoding CAZymes-and their characteristics-were compared with those of the other fungi. Homologous genes involved in triterpenoid, polysaccharide, and flavonoid biosynthesis were identified, and their expression was examined during four developmental stages, 10 and 20 days old mycelia, 1 year old and 3 years old fruiting bodies. Importantly, the lack of chalcone isomerase 1 in the flavonoid biosynthesis pathway suggested that different mechanisms were used in this mushroom to synthesize flavonoids than those used in plants. In addition, 343 transporters and 4 velvet family proteins, involved in regulation, uptake, and redistribution of secondary metabolites, were identified. Genomic analysis of this fungus provides insights into its diverse secondary metabolites, which would be beneficial for the investigation of the medical applications of these pharmacological compounds in the future.

Inonotus obliquus (Pers.:Fr.) Pilát has been traditionally used as a folk remedy for treatment of cancers, cardiovascular disease and diabetes in Russia, Poland, and most of the Baltic countries, but natural reserves of this fungus have nearly been exhausted. This study was designed to investigate the artificial cultivation of I. obliquus and the antitumor activity of its tissues. The ethanol extract of cultivated sclerotium had the highest cell growth inhibitory rate (74.6%) as determined by an 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. 78% of the bags produced sclerotia and only 6.17 g/bag of sclerotium was obtained. Extracts of the cultivated fruiting body showed 44.2% inhibitory activity against tumor cells. However, the yield was as high as 18.24 g/bag, and 98% of the bags produced fruiting body. The results of gas chromatography-mass spectroscopy (GC-MS) showed that similar compounds were extracted from the wild and cultivated samples. The principal compounds observed were lanosterol, inotodiol, and ergosterol. Their percentages of the mass fraction were 86.1, 59.9, and 71.8% of the total, for the wild sclerotium, cultivated sclerotium, and cultivated fruiting body, respectively. Ergosterol was found to be much higher (27.32%) in cultivated fruiting body. We conclude that cultivated fruiting body of I. obliquus obtained by inoculation of the substrate with spawn mycelium of the fifth generation could serve as an ideal substitute for the wild I. obliquus.

Compound (1-(4-hydroxyphenyl)-3-methoxypropan-1-one) is a newly isolated compound from Inonotus obliquus. In this paper, four target protein prediction methods were used to search the potential targets of compounds. The target proteins were determined according to the corresponding screening criteria. The target proteins with therapeutic value were docked with compound and target proproteinogen ligands by molecular docking software, and their binding affinity with target proteins was compared. The pharmacophore analysis program was used to investigate whether the pharmacophore characteristics of the compound and the original ligand compounds were consistent. 26 target proteins were obtained by virtual screening. The PASS server filtered out 2 targets. 7 targets were screened by the PharmMapper database. 2 targets were obtained by screening the PDB protein database. 18 targets were obtained by ROCS screening. Compared with the binding affinity of molecular docking, it was found that the binding affinity of the compound to oxysterols receptor LXR-beta, chymase, heat shock protein HSP 90-alpha, carbonic anhydrase 2 and arylamine N-acetyltransferase Nat was higher than that of the original ligands. It shows that the compound has potential applications in anti-tumor, anti-inflammatory, affecting intestinal flora and affecting human lipid transport. Pharmacophore analysis was performed using LigandScout software. The results of pharmacophore analysis show that the phenolic hydroxyl group in the compound is the key group and can participate in the interaction with amino acids. Besides, the mother nuclear benzene ring can also participate in the hydrophobic interaction.
