Characterization of the transcriptome profiles related to globin gene switching during in vitro erythroid maturation

Abstract: BackgroundThe fetal and adult globin genes in the human β-globin cluster on chromosome 11 are sequentially expressed to achieve normal hemoglobin switching during human development. The pharmacological induction of fetal γ-globin (HBG) to replace abnormal adult sickle βS-globin is a successful strategy to treat sickle cell disease; however the molecular mechanism of γ-gene silencing after birth is not fully understood. Therefore, we performed global gene expression profiling using primary erythroid progenitors… Show more

“…The globin family is known to play an important role in oxygen transport. Down-regulation of these genes may interfere with either erythropoiesis (which involves actively dividing hematopoietic cells), or heme biosynthesis (which is required for normal hemoglobin production; Li et al, 2012). Blockage of oxygen transport could potentially play an important role in silver ioninduced toxicity.…”

“…Expression of KLF1 mRNA in the adult mouse is restricted to the bone marrow and the red pulp of the spleen (3, 100). KLF1 transcript levels dramatically increase during hematopoietic differentiation, both in vivo and in cell culture assays (101)(102)(103). This is also reflected in the dynamic changes in histone marks (particularly H3K4/K9/ K27/K36 and H4K20) and H2A.Z and RNA Pol II occupancy (101).…”

“…Its onset in the yolk sac is strictly limited to the mesodermal, primitive erythroid cells that populate the blood islands at the early headfold stage (embryonic day 7.5 [E7.5]), switching by E9.5 to definitive cells within the hepatic primordia and then to the red pulp of the adult spleen and the bone marrow (6). During definitive hematopoietic differentiation, EKLF is expressed at low levels in multipotent progenitors (MPP) and retains an expression pattern restricted to the common myeloid progenitor (CMP) and megakaryocyte erythroid progenitor (MEP) prior to eventual segregation to erythroid progeny (7)(8)(9).…”
