“…Although there is evidence for the existence of GlyRα homomeric receptors, α/β hetero-oligomers prevail in the ventral spinal cord during adulthood (Becker et al, 1988 ; Langosch et al, 1988 ). The subunit composition, however, is still controversially discussed ranging between a 2α3β stoichiometry to a 4α1β (Grudzinska et al, 2005 ; Yu et al, 2021 ). To date, four α subunits (α1–4) and one β subunit are known, all of which are expressed in the rodent spinal cord, but only the α subunits α1- α3 and the β subunit are being expressed in humans (Dutertre et al, 2012 ).…”

“…The β Subunit Converts α2 Subunits to an "α1-Like" Glycine Receptor With Respect to Ethanol Sensitivity Glycine receptors can be expressed in recombinant systems as homomeric or heteromeric complexes (4 α subunits:1 β subunit) (Yu et al, 2021;Zhu and Gouaux, 2021) and their expression can be monitored looking at changes on their properties such as time to activation and glycine affinity (Figure 1A). Figure 1B shows current traces activated by an EC 10 concentration of glycine in homomeric and heteromeric GlyRs.…”

“…GlyR-Abs was firstly reported to be related to PERM in 2008 by Hutchinson et al ( 2 ), and they were most frequently found in patients with PERM ( 3 ). Glycine receptors are pentameric ligand-gated chloride channels present mainly in the adult brainstem and spinal cord, facilitating inhibitory signaling, and the stoichiometry has been recently proved to be 4 alpha: 1 beta subunits ( 4 ). Mutations of GlyRα1 result in hereditary hyperekplexia, a disorder characterized by excessive startle responses in infancy, which resembles the phenotype of PERM ( 5 ).…”
