Characterization of RNA from Exosomes and Other Extracellular Vesicles Isolated by a Novel Spin Column-Based Method

Abstract: Exosomes and other extracellular vesicles (commonly referred to as EVs) have generated a lot of attention for their potential applications in both diagnostics and therapeutics. The contents of these vesicles are the subject of intense research, and the relatively recent discovery of RNA inside EVs has raised interest in the biological function of these RNAs as well as their potential as biomarkers for cancer and other diseases. Traditional ultracentrifugation-based protocols to isolate EVs are labor-intensive … Show more

“…Differential ultracentrifugation is the gold standard for exosomes isolation; however, it requires specific investment, and the process is time consuming [30]. Furthermore, a limited amount of plasma results in a lower yield when subjected to repeated ultracentrifugation.…”

“…However, Exoquick reagent and Qiagen exoEasy Maxi Kit have obtained exosomes by compound polymerization precipitation or membrane affinity centrifugation columns. The operation is simple and fast, and it is suitable for extraction of serum exosomes (12,13).…”

“…It appears reasonable to approach this issue by detecting the EML4-ALK fusion transcript. To this aim, the RNA protected in vesicles as the exosomal RNA appears to be a promising source for this kind of analyses and some diagnostic tests designed to identify the predictive biomarker EML4-ALK in liquid biopsy are based on EML4-ALK fusion transcripts detection using exosomal RNA (68).…”
