Abstract: Stem cell transplantation is a promising therapeutic strategy to enhance axonal regeneration after spinal cord injury. Unrestricted somatic stem cells (USSC) isolated from human umbilical cord blood is an attractive stem cell population available at GMP grade without any ethical concerns. It has been shown that USSC transplantation into acute injured rat spinal cords leads to axonal regrowth and significant locomotor recovery, yet lacking cell replacement. Instead, USSC secrete trophic factors enhancing neurit… Show more

“…sGene, genes encoding secreted proteins; sLungGenes, genes encoding secreted proteins that are preferentially expressed in the adult lung; z-P Lung , lung-preferential expression Z score. mesenchymal stromal cells (57,58). Our analysis of the human and mouse tissue databases indicated preferential expression of Ndnf in the normal adult lung in humans (Figure 1 and Supplemental Figures 1 and 2) and mice ( Figure 1 and Supplemental Figure 3), which we validated using several approaches.…”

“…Afterward, confluent cell layers were washed twice with PBS to remove fetal bovine serum and cells were then cultured for 24 h in serum‐free N2‐medium modified according to Bottenstein and Sato (19) and Schira et al . (20) supplemented with 2 µM forskolin. Medium was changed again to exclude FBS effects and Schwann cells were incubated for 48 h in 10 ml N2‐medium supplemented with either 0.1 µM FTY720P (Echelon Biosciences, Salt Lake City, UT, USA) or control buffer as previously described (13).…”
