Characterization of Muscarinic Acetylcholine Receptors in the Rat Epididymis1

Abstract: The aim of the present study was to characterize the muscarinic acetylcholine receptor subtypes present in the caput and cauda of rat epididymis. The specific binding of [3H]quinuclidinyl benzilate ([3H]QNB) to epididymal membranes was time dependent, temperature dependent, and saturable. The cauda epididymis showed higher affinity to [3H]QNB and higher muscarinic receptor density when compared to the caput region. The [3H]QNB binding was tested in competition studies with different muscarinic receptor antagon… Show more

“…Hippocampus membrane, obtained from 6 animals for each experiment, was prepared as described by Maróstica et al [20]. Briefly, hippocampus was isolated from all experimental groups, minced and homogenized in 25 m M Tris-HCl, pH 7.4 (containing 0.3 M sucrose, 5 m M MgCl 2 , 1 m M EDTA and 1 m M phenylmethylsulfonyl fluoride) with a Ultra-Turrax homogenizer (T-25, Ika Labortechnik, Staufen, Germany).…”

“…Saturation-binding experiments were performed as previously described [20]. Briefly, hippocampus membrane preparation (80 µg protein/ml) was incubated with 0.05–8.0 n M [ 3 H]quinuclidinyl benzilate ([ 3 H]QNB) in the absence (total binding) and presence (nonspecific binding) of 1 µ M atropine for 1 h at 30°C.…”
