Low levels of genetic structuring in King George whiting Sillaginodes punctatus across two geographic regions

“…Genomic DNA was extracted from a total of 510 individuals using QIAGEN DNEasy Blood and Tissue kits (http://www.qiagen.com), as per the manufacturer's instructions. Levels of connectivity were assessed using seven polymorphic microsatellite markers: Sp19, Sp22, Sp32, Sp35, Sp36, Sp38 and Sp39 (Haigh & Donnellan, ). Microsatellites were amplified using a PCR touchdown programme using the following thermal cycling conditions: initial hot start at 94° C for 15 min; 5 cycles of 94° C for 45 s, 65° C for 45 s, 72° C for 45 s; 5 cycles of 94° C for 45 s, 60° C for 45 s, 72° C for 45 s; 10 cycles of 94° C for 45 s, 57° C for 45 s, 57° C for 45 s, 72° C for 45 s; 20 cycles of 94° C for 45 s, 55° C for 45 s, 72° C for 45 s; final elongation at 72° C for 15 min.…”
