Characterization of microbial siderophores by mass spectrometry

Abstract: Siderophores play important roles in microbial iron piracy, and are applied as infectious disease biomarkers and novel pharmaceutical drugs. Inductively coupled plasma and molecular mass spectrometry (ICP-MS) combined with high resolution separations allow characterization of siderophores in complex samples taking advantages of mass defect data filtering, tandem mass spectrometry, and iron-containing compound quantitation. The enrichment approaches used in siderophore analysis and current ICP-MS technologies a… Show more

“…For example, siderophores, which are important natural iron chelators, represent a novel class of antibiotics with considerable therapeutic potential [53]. Chitinase acts as an antagonist against fungi by inhibiting spore germination and germ-tube elongation [32].…”

“…To circumvent these problems, pre-concentration of samples with solid-phase extraction and HPLC is used to separate siderophores (Pluhacek et al, 2016). Mass spectrometry is the most important and promising method to discover and characterize siderophores and associated receptors in fully sequenced microorganisms (Lv et al, 2014;Mathavan et al, 2014;Podkowa et al, 2014) (Fig.…”

“…Specifically, different types of mass spectrometry, such as inductively coupled plasma-mass spectrometry (ICP-MS), matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS), electrospray-ionization mass spectrometry (ESI-MS), affinity selection-mass spectrometry (ASMS), laser ablation electrospray ionization-mass spectrometry (LAESI-MS), and mass spectrometry imaging (MSI) techniques (MALDI-MS imaging and DESI-MS imaging), have been broadly used to identify siderophores (Pluhacek et al, 2016;Su et al, 2016;Hagan et al, 2017;Walker et al, 2017). In short, mass spectrometry and associated technologies are emerging as novel tools for the annotation of chemical, biological, and biomedical applications of pathogen-associated siderophores.…”

“…Pyoverdines are a large family of nonribosomal peptides and siderophores (ie, iron‐chelating compounds) produced by pseudomonads . Using pyoverdine as an example, in this work, we show how our open‐source tool CycloBranch can also be used to reveal the mutual ratio of 2 siderophores in a mixture.…”

“…16 Pyoverdines are a large family of nonribosomal peptides and siderophores (ie, iron-chelating compounds) produced by pseudomonads. [17][18][19] Using pyoverdine as an example, in this work, we show how our open-source tool CycloBranch can also be used to reveal the mutual ratio of 2 siderophores in a mixture. For electrospray ionization in positive ion mode, the respective pyoverdines D and E were detected as protonated molecules at m/z 1334.59713 and 1333.61310, respectively.…”
