Characterization ofSalmonellaEnteritidis strains isolated from poultry and farm environments in Brazil

Abstract: Salmonella Enteritidis is a major causative agent of foodborne outbreaks worldwide. Using enterobacterial repetitive intergenic consensus-polymerase chain reaction (ERIC-PCR) and pulsed-field gel electrophoresis (PFGE), this study assessed the genetic relatedness, the pathogenic potential, and antimicrobial resistance in 60 strains isolated from chickens and the farm environment in Brazil between 2004 and 2010. The resulting concatenated dendrogram of the two methodologies distinguished the strains into two cl… Show more

“…Level of resistance against nalidixic acid was very much in agreement to the findings of Halimi et al, who found 53% of their Salmonella isolates resistant to nalidixic acid [43]. However, Campioni et al have reported more resistance to nalidixic acid compared to our observation which can be explained due to high level of exposure of poultry to drug used in study [44] . Nalidixic acid is a quinolone drug and resistance associated with it has been due to various point mutations in DNA gyrase enzyme where the drug acts [45] .…”

“…Genomic DNA extraction of the 92 strains listed in Table 1 and determination of their concentrations were done using the methods described by Campioni et al (2014). The general PCR procedure was performed according to the method of Falcão et al (2006).…”

“…The genomic DNAs of the 92 strains were extracted as described by Campioni et al (2014). A volume of 50 ml PCR mixture was prepared for each isolate, containing primers ERIC 1 (59-ATG-TAAGCTCCTGGGGATTCAC-39) and ERIC 2 (59-AAGTAAGTGA-CTGGGGTGAGCG-39) described by Versalovic et al (1991).…”

“…The primers used for ERIC-PCR were: ERIC-1R (5′ -ATGTAAGCTCCTGG-GGATTCAC-3′ ) and ERIC-2 (5′ -AAGTAAGTGACTGGGGTGAGCG-3′ ), as previously described (Hulton et al, 1991;Campioni et al, 2014). The PCR mixture (20 μL) consisted of the following: 10 μL of One Taq 2× Master with Standard Buffer (New England Biolabs, MA, USA), 0.4 μL of 10 μM of each primer, 2 μL of DNA template, and 7.2 μL of ddH 2 O.…”
