Characterization of cloned rat ribosomal DNA fragments

Abstract: Two Charon 4A lambda bacteriophage clones were characterized which contain all and part o the 18S ribosomal DNA of the rat. One clone contained two Eco RI fragments which include the whole 18S ribosomal RNA region and part of 28S ribosomal RNA region. The other clone contained an Eco RI fragment which covers part of 18S ribosomal RNA region. There were differences between the two clones in the non-transcribed spacer regions suggesting that there is heterogeneity in the non-transcribed spacer regions of rat rib… Show more

“…Restriction maps for other rat rDNA clones (also from Sprague-Dawley libraries) have been published (4,27,32,33) and, in general, agree with our proposed map (Figure 1). One difference among the cloned rDNAs from different laboratories is the estimated size of the Eco RI fragment containing the initiation site.…”

“…Drs. Richard Torczynski and Harris Busch (Wadley Institutes of Molecular Medicine, Dallas, TX) generously provided the rat ribosomal 18s RNA probe; the probe (pDF8) contained 0.9 kb of the middle portion of the 18s DNA (Torczynski et al, 1983) subcloned into pBR322 (Fuke et al, 1981). The v-ruPu cDNA probe was obtained from the American Type Culture Collection (ATCC No.…”
