Characterization of CD1e, a Third Type of CD1 Molecule Expressed in Dendritic Cells

Abstract: Dendritic cells express several alternatively spliced CD1e mRNAs. These molecules encode proteins characterized by the presence of either one, two, or three ␣ domains and either a 51-or 63-amino acid cytoplasmic domain. Moreover, mRNAs encoding isoforms lacking the transmembrane domain are observed. Several of these CD1e isoforms were expressed in transfected cells, and two of them, with three ␣ domains, displayed a particular processing pathway. These latter isoforms slowly leave the endoplasmic reticulum due… Show more

“…S4) and partially colocalized with the Golgi marker TGN-38 and with MHC class II compartment (Fig. S3B), thus showing a distribution similar to the one occurring in human cells (1). Number and phenotype of iNKT cells in thymus and periphery as well as of other lymphocyte populations and monocytes were comparable in Tg and WT mice (Figs.…”

“…CD1e is the only CD1 molecule that becomes soluble and is never expressed on the plasma membrane, thus it cannot behave as an antigen-presenting molecule. Moreover, CD1e shows a very peculiar intracellular distribution, which in dendritic cells (DCs) correlates with their maturation stage (1). In immature DCs, CD1e prevalently accumulates in the trans-Golgi network (TGN), whereas, in mature DCs, it distributes in late endosomes and lysosomes (LYs) where it is cleaved into an active soluble form (2).…”

“…In contrast, CD1e is not expressed on the cell surface and is an intermediate form in terms of its homology to the other CD1 molecules. Accordingly, it is regarded as a group 3 CD1 molecule [3].By trafficking through endosomal compartments, CD1 proteins pick up antigens in a similar fashion to MHC class II molecules [4]. However, instead of peptides, CD1 molecules present lipids, glycolipids or lipopeptides of either foreign or self origin [2,5].…”
