Characterisation of a newly detected bacteriophage infecting Bordetella bronchiseptica in swine

Abstract: A novel virulent bacteriophage, vB_BbrM_PHB04, infecting Bordetella bronchiseptica was isolated from wastewater collected at a swine farm in China. Phage vB_BbrM_PHB04 exhibited growth over a wide range of temperature and pH conditions and showed different efficiency of plating values and lytic spectra within the same strains at 25 °C and 37 °C. High-throughput sequencing revealed that vB_BbrM_PHB04 has a linear double-stranded DNA genome with 124 putative open reading frames. Overall, the genome of vB_BbrM_PH… Show more

“…Phage isolation and purification were carried out using a doublelayer agar method as described previously (Chen et al, 2019a). Briefly, sewage samples (20 ml each) were collected in Jingzhou, China, and sterilized by filtration through a 0.22-µm membrane.…”

“…To identify strains lysogenized by PHB09, 100 µl of exponentialphase B. bronchiseptica Bb01 culture (∼10 8 CFU/ml) were incubated with 100 µl of purified phage PHB09 (10 9 PFU/ml) as previously described (CLSI, 2018;Chen et al, 2019a). Six milliliters soft TSA (0.75% agar) containing 10% fetal bovine serum was then added to the mixture and then poured onto a prepared TSA plate.…”

“…Currently, it is a well-recognized etiological agent in many domestic and wild animal diseases including tracheobronchitis, conjunctivitis, rhinitis, and pneumonia (Garbal et al 2016;Petrovic et al 2017). In particular, it is one of the primary causative agents of atrophic rhinitis and bronchopneumonia in swine (Moore and Kern 2001;Prüller et al 2015;Chen et al 2019). The characteristic lesion of atrophic rhinitis is atrophy of the nasal septum and turbinate, leading to facial deformation (Petrovic et al 2017;De Jong and Nielsen 1990;Brockmeier et al 2002).…”
