Changes in Notch signaling coordinates maintenance and differentiation of the Drosophila larval optic lobe neuroepithelia

Abstract: A dynamic balance between stem cell maintenance and differentiation paces generation of post-mitotic progeny during normal development and maintenance of homeostasis. Recent studies show that Notch plays a key role in regulating the identity of neuroepithelial stem cells, which generate terminally differentiated neurons that populate the adult optic lobe via the intermediate progenitor cell type called neuroblast. Thus, understanding how Notch controls neuroepithelial cell maintenance and neuroblast formation … Show more

“…E(spl)md, E(spl)m7, and Tom are direct targets of the Notch pathway (Bray 2006), while H is a transcription corepressor involved in repression of Notch signaling activity (Bang et al 1995). The Notch pathway is required for neuroepithelial maintenance and expansion (Egger et al 2010;Ngo et al 2010;Reddy et al 2010;Yasugi et al 2010;Orihara-Ono et al 2011;Wang et al 2011b;Weng et al 2012). Our microarray data support a model in which JAK/STAT acts upstream of Notch signaling in the optic lobe (Wang et al 2011a).…”

“…Another example of the association of neurogenesis with a particular cell cycle stage in Drosophila can be found in the neural precursors from the outer optic anlagen. This structure gives rise to the outer part of the medulla and lamina of the optic lobes, and their constituting neuroepithelial cells require to be in G1 to progress from neuroepithelial cells to neuroblasts (Reddy et al, 2010), concomitant with an increase of Notch activity that facilitates the neuroblast fate selection (Weng et al, 2012). Forcing cell cycle progression in the neuroepithelial cells with cell cycle regulators such as cyclin D, cdk4, E2F1 and DP has been shown to lead to a differentiation delay with a concomitant prevention of Dl expression (Reddy et al, 2010).…”

“…3C-F9); it started to be highly expressed at the transition zone where the Notch ligand Dl is enriched in a characteristic punctuated pattern (Fig. 3C-D0) (Weng et al, 2012;Yasugi et al, 2010). L9sc is also very locally activated at the transition zone, in progenitor I (PI) and progenitor II (PII) cells; the latter juxtaposed to the emerging medulla NBs (Yasugi et al, 2010) ( Fig.…”

“…Notch signaling is required to regulate the progression of the proneural wave (Egger et al, 2010;Ngo et al, 2010;Reddy et al, 2010;Wang et al, 2011;Weng et al, 2012;Yasugi et al, 2010). Given that Cno and Notch functionally interact in other systems (Carmena et al, 2006;Miyamoto et al, 1995) and that cno mutant phenotypes were very reminiscent of those previously described for Notch mutants in the neuroepithelium (Yasugi et al, 2010), we decided to analyze the relative localization of Cno and Notch in the optic lobe.…”
