Cell death induced by zinc and cadmium is mediated by clusterin in cultured mouse seminiferous tubules

Abstract: Sertoli cells, lining the walls of the seminiferous tubules, are in close contact with and regulate all aspects of the development of the germ cells. Clusterin, is a glycoprotein produced abundantly by Sertoli cells, and associated with either apoptosis or cell survival. Zinc is present in high concentrations in the testis and required for sperm development by an as yet unknown mechanism. Permeation of zinc into cells via voltage-gated calcium channels (VGCCs), however, is suggested to induce cell death. We ex… Show more

“…Rat SeT were used for culture instead of individual cell types, since this model has been shown to be suitable to mimic the testicular cellular environment ex vivo by several groups (Gilleron et al 2009, Kaisman-Elbaz et al 2009, GeoffroySiraudin et al 2010. Testes (90-day-old rats) were removed, trimmed free of fat, washed in cold PBS and placed in DMEM-F12 culture medium at 32 8C.…”

“…This raises the possibility that elevated sCLU levels may enhance tumorigenesis by interfering with Bax proapoptotic activities. On the other hand, several reports suggest that the nuclear form of CLU (nCLU) is a proapoptotic protein (13)(14)(15). Recently, we and others have demonstrated that CLU intracellular forms are involved in NF-B pathway regulation through IB-␣ stabilization (16,17).…”

“…Of particular importance is the activation of mitogen-activated protein kinase (MAPK) 2 and PI3 kinase pathways, leading to enhanced cell proliferation and survival (15)(16)(17)(18)(19)(20). In epidermal tissues, higher levels of zinc have been correlated with higher mitotic activity (21).…”
