Abstract: The first lineage decision during mouse development is the establishment of trophectoderm and inner cell mass lineages, morphologically distinguishable at the blastocyst stage. The Caudal-like transcription factor Cdx2 is required for repression of inner cell mass genes Oct4 and Nanog in the trophectoderm. Expression of Cdx2 in the trophectoderm is thus one of the earliest known events in lineage determination. However, it is not clear whether the Cdx2 expression pattern is the cause or the consequence of this… Show more

“…Differentiation of ES cells to a trophoblast fate can also be induced by overexpression of the caudal-type homeobox gene, Cdx2 (Niwa et al, 2005;Tolkunova et al, 2006). In early mouse embryos, loss of Cdx2 activity results in the loss of trophectodermal characteristics and acquisition of ICM character (Ralston and Rossant, 2008). Cdx2 has been shown to be required for the repression of the ICM genes, Oct3/4 and Nanog.…”

“…Thus, certain features of TE, i.e., cavity expansion and hatching, were enhanced in ESCM, whereas the other feature, i.e., Cdx2 expression, was not. Interestingly, Cdx2 is important for the differentiation of trophoblast to enable implantation and placenta formation, but it is dispensable for the blastocyst cavity formation [36,77]. Thus, it is possible that the impact of ESCM on blastocyst development may be more selective than generally speeding up development.…”
