CD56bright NK cells after hematopoietic stem cell transplantation are activated mature NK cells that expand in patients with low numbers of T cells

Abstract: We studied early NK-cell recovery in 29 allografted patients undergoing different lymphoreductive regimens. Already at 2 wk after graft take, the number of NK cells had reached (supra)normal levels but NK-cell subsets were skewed. The number of CD56 dim CD16 bright NK cells was low and correlated strongly with the level of hematopoiesis, whereas the number of the more abundant NK cells expressing high levels of CD56 did not. Post-transplant CD56 bright NK cells (ptCD56 bright ) differed from CD56 bright NK cel… Show more

“…Finally, NK cell reconstitution was similar in both arms, with faster recovery of CD56 bright NK cells in comparison with CD56 dim NK cells, as reported previously by other groups of investigators (49) and particularly after HLA-haploidentical stem cell transplantation (50).…”

“…NK cell subset distribution is significantly altered after HSCT with an overrepresentation of CD56 bright cells and a reduction in fully differentiated, cytotoxic CD56 dim cells (18,19). Furthermore, an intermediately differentiated CD16 + CD56 bright NK subset (Fig.…”

“…Early after an HCT, CD56 bright NK cells are the major lymphocyte subset to recover in the peripheral blood (73)(74)(75)(76)(77)(78). Functional CD56 bright NK cell antileukemic responses have not been analyzed in depth in this setting (57).…”

“…Functional CD56 bright NK cell antileukemic responses have not been analyzed in depth in this setting (57). In 2 reports, an "activated" phenotype with an intact CD56 bright response to combined cytokine activation was observed (76,78), although responses to leukemia or other target cells were not assessed. In a more recent study focused on KIR + CD56 dim NK cells from HCT recipients, IL-15 stimulation led to a significant enhancement of the antileukemia responses of CD56 dim NK cells; however, the effects of IL-15 on CD56 bright NK cell responses to leukemia cells were not reported (77).…”
